摘要
以‘穆拉诺’(Murano)、‘孟菲斯’(Memphis)、‘凯尔索’(Kelso)、‘刀锋’(Blade)、‘色拉达’(Salada) 5个品种彩色马蹄莲的块茎为试验材料,对其进行灭菌时间、灭菌方法及不定芽诱导条件的筛选。结果表明,在灭菌时间上,不同品种间存在显著性差异,‘穆拉诺’、‘凯尔索’、‘刀锋’的最佳灭菌时间是15 min,‘孟菲斯’、‘色拉达’的最佳灭菌时间是18 min;灭菌方法,5个品种均在45℃水浴1 h和杀菌剂500倍液处理4 h的条件下,灭菌效果最好,初代培养成活率分别达到50%和70%左右;不定芽诱导阶段,不同品种间存在显著性差异,‘穆拉诺’、‘孟菲斯’、‘凯尔索’、‘色拉达’的最适培养基为MS+2 mg/L 6-BA+0.1 mg/L NAA,‘刀锋’最适培养基为MS+3 mg/L 6-BA+0.1 mg/L NAA。
        The tubers of five species of Zabtadeschia hybrida, including 'Murano', 'Memphis', 'Kelso', 'Blade' and'Salada', were used as test materials to select the sterilization time, sterilization method and the adventitious bud induction conditions. The results showed that for the time of sterilization, there were significant differences between different varieties. The optimal sterilization time of 'Murano', 'Kelso' and 'Blade' was 15 min, and the best sterilization time of 'Memphis' and 'Salada' was 18 min. For sterilization method, five varieties were all under the condition of 45℃ water bath for 1 h and 500 times of bactericide for 4 h, and the sterilization effect was the best that original culture survival rate reached 50% and 70%, respectively. At the adventitious bud induction phase, there was a significant difference between different varieties. The optimal culture medium for 'Murano', 'Memphis', 'Kelso', and'Salada' was MS +2 mg/L 6-BA +0.1 mg/L NAA, and the optimal culture medium for 'Blade' was MS +3 mg/L6-BA+0.1 mg/L NAA.
