摘要
目的:优化金银花的提取工艺,建立高效液相色谱法(HPLC)快捷检测绿原酸和异绿原酸A的方法。方法:以HPLC同时测定绿原酸、异绿原酸A的含量为指标,通过比较醇浓度、回流时间、超声功率、超声时间4因素对提取工艺的影响,优化超声波辅助醇法提取金银花的工艺。结果:以绿原酸、异绿原酸A的综合评分为指标,最佳提取工艺是60%乙醇超声(功率500 W)45 min,再回流提取1 h。结论:建立了准确灵敏的检测方法,优化的提取工艺经济、高效,能为生产提供理论依据。
Objective:To optimize the extraction process of Lonicerae flos and to establish an HPLC method to detect chlorogenic acid and isochlorogenic acid A rapidly.Methods:The content of chlorogenic acid and isochlorogenic acid A was simultaneously determined by HPLC and used as an indicator to compare the effect of the four factors(alcohol concentration,reflux time,ultrasonic power and ultrasonic time) on the extraction process,optimize the extraction process of Lonicerae flos by ultrasonic-assisted alcohol method.Results:Taking the comprehensive score of chlorogenic acid and isochlorogenic acid A as an indicator,the best extraction process was using 60% alcohol ultrasounding(power 500 W) for 45 min,then reflux extraction for 1 hour.Conclusions:An accurate and sensitive detection method is established,the optimized extraction process is economical and efficient,which can provide theoretical basis for production.
