摘要
目的:研究谷胱甘肽硫转移酶基因GSTT1、GSTM1以及多药耐药基因MDR1多态性与儿童急性淋巴细胞白血病(ALL)化疗反应的关系。方法:筛选ALL患者100例,采用多重PCR技术分析GSTT1、GSTM1以及MDR1基因型,比较不同基因型患者化疗反应的差异。结果:GSTT1、GSTM1以及MDR1 3种基因非缺失型个体发生ALL的风险明显高于GSTT1、GSTM1以及MDR1任一基因缺失型,GSTT1、GSTM1以及MDR1 3种基因缺失型患者CR率较GSTT1、GSTM1以及MDR1基因非缺失型患者低,差异均有统计学意义(P<0.05)。结论:GSTT1、GSTM1以及MDR1基因缺失型个体对ALL的易感性升高,化疗完全缓解率显著降低。GSTT1、GSTM1以及MDR1基因型检查有助于指导ALL儿童患者个体化治疗方案的制定。
        Objective:To investigate the impact of GSTT1,GSTM1and MDR1genotypes on chemotherapy response of childhood acute lymphoblastic leukemia(ALL).Methods:GSTT1,GSTM1and MDR1genotypes were analyzed with multi-PCR in 100ALL patients.Results:The risk of ALL in children carrying GSTT1,GSTM1,and MDR1null genotypes was significantly higher than in those carrying GSTT1,GSTM1,and MDR1presence genotypes,the CR ratio in children carrying GSTT1,GSTM1,and MDR1null genotypes was significantly lower than n those carrying GSTT1,GSTM1,and MDR1presence genotypes,there was statistical significance(P<0.05).Conclusions:Children carrying GSTT1,GSTM1and MDR1null genotypes have a significantly higher risk of suffering from ALL and a significantly lower chemotherapy CR ratio than those who carry GSTT1,GSTM1and MDR1presence genotypes.GSTT1,GSTM1and MDR1genotypes test might be useful in selecting appropriate chemotherapy regimens for patients with ALL.
