脏腑图点穴法对胃肠动力障碍大鼠Cajal间质细胞及抑制性神经递质的影响

摘要
目的观察脏腑图点穴法对胃肠动力障碍大鼠Cajal间质细胞(ICC)及NO、NOS的影响,探究脏腑图点穴法影响胃肠动力的可能机制。方法 32只SD大鼠随机分为空白组、模型组、点穴组、药物组。除空白组外,均用多因素联合造模法建立功能性消化不良(FD)大鼠模型。造模成功后各组予相应干预,持续14天。测量各组大鼠胃内残留率,并取胃窦组织检测c-kit受体表达、一氧化氮(NO)含量、一氧化氮合酶(NOS)活性。结果脏腑图点穴法可以降低FD大鼠胃内残留率(P<0.05);提高c-kit蛋白的表达(P<0.05);降低抑制性神经递质合成酶诱导型NOS(i NOS)活性(P<0.05)。组织总NOS(t NOS)活性及NO含量有降低趋势,但差异无统计学意义。结论脏腑图点穴法可调节FD大鼠胃肠动力,该作用可能与其改变抑制性神经元上NOS活性及NO释放,进而促使ICC修复有关。
Objective To observe the influence of acupoint pressure from zang-fu diagram method on Cajal mesenchymal cells( ICC) and NOS( NOS) in rats with gastrointestinal dynamic disorder,and to explore the possible mechanism of the influence of acupoint pressure from zang-fu diagram method on gastrointestinal dynamics. Methods 32 SD rats were randomly divided into blank group,model group,acupoint pressure group,and drug group. In addition to the blank group,the functional dyspepsia( FD) rat model was established by multifactor joint molding method. After successful modeling,each group was given corresponding intervention for 14 days. The gastric residual rate of rats was measured and the expression of c-kit receptor,nitric oxide( NO) and nitric oxide synthase( NOS) activity were detected. Results The residual rate( P < 0. 05) was reduced in rats with the acupoint pressure from zang-fu diagram. The expression of c-kit protein was increase( P < 0. 05). The activity of NOS( i NOS) induced by inhibitory neurotransmitter synthesis was reduced( P < 0. 05). The activity of total NOS( t NOS) and the content of NO were decreased,but there was not statistically significant. Conclusion The acupoint pressure from zangfu diagram method can modulate the gastrointestinal motility of FD rats,which may change the NOS activity and NO release of the inhibitory neurons,and then to promote the ICC repair.
