摘要
研究显齿蛇葡萄中不同黄酮组成对α-葡萄糖苷酶的抑制活性及DPPH·清除能力。15份不同显齿蛇葡萄叶,甲醇超声辅助法制备供试品;α-葡萄糖苷酶抑制和DPPH·清除模型评价相关生物活性;拟复方验证原样品的相应生物活性。结果表明:对α-葡萄糖苷酶抑制活性最好的植株为ZF3号, IC_(50)为0.007 mg/mL;α-葡萄糖苷酶IC_(50)与二氢杨梅素、杨梅素含量呈极显著正相关(p<0.01),相关系数分别为0.724, 0.870;与杨梅苷含量呈显著负相关(p<0.05),相关系数为0.604。拟复方样品的活性与之对应原植物样品相当,但略低于原样品。显齿蛇葡萄叶中二氢杨梅素、杨梅素和杨梅苷对其α-葡萄糖苷酶的抑制活性有显著影响,而对DPPH·清除能力影响不显著;二氢杨梅素含量低的显齿蛇葡萄叶适宜开发降糖产品。
        Inhibition of flavonoids composition on α-glucosidase activity and DPPH· clearance in Ampelopsis grossedebtata grape. 15 kinds of grape leaves of Ampelopsis grossedebtata with different contents of dihydromyricetin, myricetin and myricitrin, prepared by ultrasonic assisted extraction of methanol; Evaluation of related biological activities by α-glucosidase inhibition model and DPPH· clearance model; Verification of the bioactivity of the original sample by compound method(according to the ratio of the three flavonoids in the sample). The results showed the α-glucosidase inhibitory activity and DPPH· scavenging ability of different flavonoids from Ampelopsis grossedebtata leaf extract. The contents of dihydromyricetin and myricetin were significantly positively correlated with their α-glucosidase IC_(50)(p<0.01), and the correlation coefficients were 0.724 and 0.870, respectively. The content of myricitrin was negatively correlated with α-glucosidase IC_(50)(p<0.05), and the correlation coefficient was 0.604. However, the correlation between the three flavonoids and DPPH· scavenging ability was weak and the effect was small. The activity of the compound sample was similar to that of the original plant sample, but slightly lower than the original sample. Dihydromyricetin, myricetin and myricitrin in Ampelopsis grossedebtata leaves had significant effects on the α-glucosidase inhibitory activity, but had no significant effect on DPPH· scavenging ability. The Ampelopsis grossedebtata leaves with low dihydromyricetin content were suitable for development of hypoglycemic products.
