摘要
Wnt2蛋白是Wnts蛋白家族的重要成员,参与卵巢的发育。本研究以NCBI蛋白质数据库中发布的Wnt2蛋白氨基酸序列为研究对象,采用生物信息学软件对Wnt2蛋白的理化性质、信号肽及跨膜结构域、糖基化和磷酸化位点、二级结构和功能结构域、亚细胞定位和功能结构分类、序列相似比对及聚类、三级结构进行预测分析。结果发现,绵羊Wnt2由360个氨基酸组成,其等电点为9.21,有糖基化位点和磷酸化位点,是一个有信号肽的稳定的蛋白;二级结构与三级结构一致,均显示Wnt2由α-螺旋、延伸链、无规则卷曲构成;绵羊Wnt2蛋白是细胞外蛋白,主要参与信号转导和转录调控。研究结果为深入探讨Wnt2基因及其编码蛋白的结构和功能提供相关依据。
        Wnt2 protein is an important member of Wnts protein family and is involved in the development of ovarian. In this study, the amino acid sequence of Wnt2 protein published in NCBI protein database was used as the research object. Bioinformatics software was used to predict and analyze the physicochemical property,signal peptide and transmembrane region, glycosylation and phosphorylation site, secondary structure and functional domain, subcellular localization and functional category, sequence similarity comparison and clustering,tertiary structure of Wnt2 protein. The results showed that the Wnt2 protein in sheep was a stable protein with signal peptide, composed of 360 amino acids and with glycosylation site and phosphorylation site. The isoelectric point was 9.21. The secondary structure was composed of α helix, extended chain and irregular coil, and the tertiary structure was basically consistent with it. Wnt2 protein in sheep was extracellular protein, involved in signal transduction and transcription regulation. These results would provide theoretical basis for further study on Wnt2 gene and the structure and function of encoded protein.
