摘要
目的:确定余甘子药材的最佳提取工艺条件。方法:以没食子酸、没食子儿茶素、表儿茶素、柯里拉京、鞣花酸5个成分含量为考察指标,采用正交设计试验分别考察了药材料液比、提取时间和提取次数对超声提取、回流提取及减压提取的影响,得到3种提取方法的最佳工艺,并基于抗DPPH氧化活性进行3种最佳提取工艺的生物活性比较。结果:余甘子超声提取的最佳工艺为加入8倍量水,超声3次,每次20min;常压回流提取的最佳提取工艺为加入12倍量水,提取2次,每次1h。减压回流提取的最佳提取工艺为加8倍量水,提取3次,每次2h。且常压回流提取物具有最优的抗DPPH氧化活性。结论:3种最佳提取工艺均具有好的提取效果,但相比超声提取,常压回流与减压回流提取的提取物抗氧化活性更高,因此回流提取更加适合余甘子药材的提取。
Objective: To determine the optimum extraction conditions of Phyllanthis Fructus. Methods: The content of 5 components of gallic acid, gallic catechins, epicatechin, coriagin and tannic acid were taken as indexes. Orthogonal design was used to investigate the effects of the ratio of liquid to drug, extraction time and extraction times on ultrasonic extraction, atmospheric reflux extraction and decompression reflux extraction. The best process of three extraction methods was obtained and the bioactivity of the three best extraction processes based on the anti DPPH oxidation activity was compared. Results: The optimum ultrasonic extraction of Phyllanthus emblica was: adding 8 times of water, three times of ultrasound, 20 minutes each time; the optimum extraction process of atmospheric reflux extraction was adding 12 times of water, extracting twice, 1 hour each time. The best optimum extraction process of decompressing reflux extraction was adding 8 times of water, extracting 3 times, 2 hours each time. And the atmospheric reflux extraction had best anti DPPH oxidation activity. Conclusion: Three of the optimum extraction process obtained has good extraction effect, but compared with ultrasonic extraction, atmospheric reflux extraction and decompression reflux extraction have higher antioxidant activity, so the reflux extraction is more suitable for Phyllanthis Fructus.
