烙灸对兔膝骨性关节炎软骨细胞外基质Ⅱ型胶原、蛋白多糖表达的影响

摘要
目的:观察烙灸疗法对兔膝骨性关节炎(KOA)软骨细胞外基质Ⅱ型胶原、蛋白多糖蛋白表达的影响。方法:选取成年健康新西兰兔30只随机分为正常对照组(10只)、造模组(20只)。采用Hulth法复制兔膝骨性关节炎模型后随机分为模型对照组(10只)和烙灸干预组(10只)。烙灸干预组给以兔左后膝关节局部烙灸治疗,1次/天,20 min/次,连续4周。疗程结束后,取动物标本进行大体形态学和HE染色病理学检测,采用蛋白免疫印迹法检测软骨Ⅱ型胶原和蛋白多糖。结果:烙灸干预组软骨组织的改良Mankin's评分、软骨外基质的Ⅱ型胶原和蛋白多糖的蛋白表达均较模型对照组降低(P <0. 05)。结论:烙灸疗法可以抑制软骨细胞外基质Ⅱ型胶原和蛋白多糖降解,改善外基质环境,减少兔KOA关节软骨的破坏。
        Objective: To observe the effect of cauterization moxibustion on the expressions of collagen type Ⅱ and proteoglycan of cartilaginous extracellular matrix in rabbits with knee osteoarthritis( KOA). Methods: 30 adult healthy New Zealand rabbits were randomly divided into the blank control group( n = 10),the model group( n = 10),and the cauterization moxibustion group( n = 10). KOA model was established by Hulth method.The cauterization moxibustion was intervened with local cauterization moxibustion on the left posterior knee joint. The cauterization was once per day,20 min per time for four weeks. At the end of the treatment,the gross morphology and HE staining pathology of the animal specimens were detected,and the collagen type Ⅱ and proteoglycan of cartilage were detected by Western blot method. Results: The modified Mankin's score of cartilage tissue,the expressions of collagen Ⅱ and proteoglycan were significantly decreased in the cauterization group than those in the model group( P < 0. 05). Conclusion: Cauterization moxibustion can inhibit the degradation of collagen type Ⅱ and proteoglycan,it can improve the environment of extracellular matrix and reduce the destruction of articular cartilage in KOA rabbits.
