摘要
【目的】为了探明迷迭香精油在体内外试验中的抗菌作用。【方法】采用药敏试验检测迷迭香精油体外抑菌效果;通过金黄色葡萄球菌(金葡菌)滴鼻法建立小鼠肺炎模型,小鼠每天2次吸入迷迭香精油,每次30分连续7d,试验结束小鼠称重、采血,生化分析仪分析血常规指标,组织固定制备石蜡切片,显微镜观察组织病变,细菌定殖检测。【结果】迷迭香精油组大肠杆菌、金葡菌和链球菌抑菌圈分别为(12.4±2.4)、(12.6±1.2)、(11.5±0.5)mm,茶树精油有抑菌圈出现,薰衣草精油组无抑菌圈。模型组小鼠金葡菌感染后反应明显增强、采食减少、体重明显减轻,肺指数升高,肺细菌定殖率高,白介素-6 (IL-6)和炎性细胞数量明显增加;肉眼观察到小鼠肺脏肿胀充血、色泽暗红,肺叶上均有大小不等的出血斑,肺组织有硬肿块,肺门淋巴结肿大;镜检发现肺脏结构改变明显,肺泡间隙有炎性细胞聚集和浸润,组织实变明显,肺泡壁增厚,肺泡组织结构完整性破坏,肺泡腔内可见出血。与模型组比较,迷迭香组小鼠反应温和,采食和体重增加,肺脏指数降低和细菌定植率下降,后期IL-6和炎性细胞数量明显减少;小鼠肺组织充血减轻,肺泡间隙炎性细胞浸润减少,肺泡结构完整性逐步恢复,接近对照组小鼠。【结论】迷迭香精油体外具有良好的抗菌作用,体内缓解金葡菌引起的肺炎炎症症状。
        【Objective】It is aimed to investigate the antibacterial effects of rosemary essential oil by both in vivo and in vitro experiments.【Methods】The antibacterial effects of rosemary essential oil in vitro were detected by drug susceptibility test.Mouse pneumonia model was established by staphylococcus aureus nasal drip.Then the mice smell rosemary essential oil twice a day,30 min every time,for seven consecutive days.Tissue sections were prepared by tissue fixation,tissue lesions were observed under microscope,and bacterial colonization was also detected.【Results】The E.coli,Staphylococcus aureus and Streptococcus in the rosemary essential oil group had obvious inhibition zones of 12.4±2.4,12.6±1.2 and 11.5±0.5(mm),respectively.The tea tree oil had an inhibition zone,but the lavender group did not.In the model group,the response of mice with staphylococcus aureus infection was significantly increased,but the amount of IL-6 and inflammatory cells significantly increased.Their lung looked likes welling,hyperemia and dark red color.There were blood spots of different sizes on the lungs,hard masses in the lung tissue and enlarged hilar lymph nodes.The obvious changes in lung structure,inflammatory cell aggregation and infiltration in the alveolar space were observed by microscope,as well as obvious tissue consolidation,thickening of alveolar wall,destruction of alveolar structure integrity,and bleeding in alveolar cavity.Compared with the model group,the mice in rosemary group showed mild reaction,food intake and bodyweight increasing,and decreased their lung index and bacterial colonization rate,and significantly decreased IL-6 and inflammatory cells in the later stage.There was a decrease in lung tissue hyperemia in mice,inflammatory cell infiltration in alveolar space,and a gradually recovery in alveolar structural integrity which was close to those in the control one.【Conclusion】Rosemary essential oil has a good antibacterial effect in vitro and can relieve the inflammation of the mice pneumonia caused by staphylococcus aureus in vivo.
