摘要
目的建立采用高效液相色谱法(HPLC)测定杜仲平压片中槲皮素的含量。方法色谱柱为Thermo Hypersil Gold-C18(4.6 mm 250 mm,5μm),流动相为乙腈-0.4磷酸溶液(27∶73),流速1.0 mL·min ~(-1),柱温35℃,检测波长为256 nm。结果槲皮素在3.21~45.02μg·mL~(-1)范围内线性关系良好(R~2=0.9999),平均回收率为96.83%(RSD=1.11%)。结论所建立的方法结果准确度高,专属性强,重现性好,可用于测定杜仲平压片中槲皮素的含量。
Objective To establish High Performance Liquid Chromatography(HPLC) method to determine quercetin in Duzhong pingya Tablet. Methods The chromatographic column was Thermo Hypersil Gold-C18(4.6 mm×250 mm,5 μm),and the mobile phase was consisted of acetonitrile and 0.5% phosphoric acid(27∶73).The flow rate was 1.0 mL·min~(-1). The column temperature was set at 35 ℃, and the detection wavelengths were 256 nm. Results The linearity of chlorogenic quercetin was good in the range of 3.21~45.02 μg·mL~(-1)(R~2=0.9999)and the average recoveries was 96.83%(RSD=1.11%)for quercetin. Conclusion The established method is accurate and reproducible,and can be applied for the determination of quercetin in Duzhong Pingya Tablet.
