大黄对瘢痕成纤维细胞增殖抑制作用及其有效成分研究

摘要
目的探讨大黄有效成分对瘢痕成纤维细胞的增殖作用。方法取中药大黄,分别以水、乙酸乙酯、石油醚、饱和正丁醇、氯仿回流提取,回流提取,得到相应提取物,以此培养人增生性瘢痕成纤维细胞,酶标仪测定增殖抑制率。以甲醇溶解具有较强增殖抑制作用的乙酸乙酯提取物,以高效液色相谱法鉴定高活性成分,同法进行抗增殖作用分析。结果大黄乙酸乙酯提取物HSF增殖抑制率17.83%,抗细胞增殖活性明显高于其他提取物及对照组,其主要活性成分为及对应HSF增殖抑制率分别为没食子酸41.76%、大黄酸36.7%、大黄素38.3%。结论大黄乙酸乙酯提取物含有活性成分没食子酸、大黄酸和大黄素,具有良好的抑制瘢痕成纤维细胞增殖作用,可作为大黄治疗增殖性瘢痕物质理论依据参考。
        Objective To investigate the effect of rhubarb active ingredient on the proliferation of scar fibroblasts.Methods The traditional Chinese medicine rhubarb was extracted with water, ethyl acetate, petroleum ether, saturated n-butanol and chloroform, and refluxed to obtain the corresponding extracts. The human hypertrophic scar fibroblasts were cultured and the proliferation inhibition was measured by microplate reader. The ethyl acetate extract with strong proliferation inhibition effect was dissolved in methanol, and the high active ingredient was identified by high performance liquid chromatography. The anti-proliferation effect was analyzed by the same method. Results The inhibition rate of HSF in ethyl acetate extract of Rhubarb was 17.83%. The anti-cell proliferation activity was significantly higher than that of other extracts and control group. The main active ingredients and corresponding HSF proliferation inhibition rates were 41.76% for gallic acid and 36.7% for rhein and emodin 38.3%. Conclusion Ethyl acetate extract of Rhubarb contains active ingredients gallic acid, rhein and emodin, which has a good effect on inhibiting the proliferation of scar fibroblasts. It can be used as a theoretical basis for the treatment of proliferative scars in rhubarb.
