摘要
该研究对卤化II型聚酮类抗生素zunyimycins产生菌链霉菌(Streptomyces sp.)FJS31-2的次级代谢产物进行挖掘。次级代谢产物经乙酸乙酯萃取浓缩后,依次采用硅胶柱层析法、薄层层析(TLC)法、高效液相色谱(HPLC)法等方法进行分离和纯化,通过质谱(MS)法和核磁共振(NMR)等分析单体化合物的结构,并采用滤纸片法对其抗菌活性进行测试。结果表明,初次从该菌发酵物中分离得到多环内酯类化合物UK-78623,其对小肠结肠炎耶尔森氏菌(Yersinia enterocolitica)具有明显抑制作用,其抑菌圈直径为25.2 mm。
The secondary metabolites of Streptomyces sp. FJS31-2 producing halogenated type II polyketide antibiotics zunyimycins were further excavated. The secondary metabolite was extracted and concentrated by ethyl acetate, then isolated and purified by silica gel column chromatography,thin layer chromatography(TLC), HPLC and other methods. The structure of monomeric compound was analyzed by mass spectrometry and nuclear magnetic resonance method, and the antibacterial activity of the monomeric compound was determined by paper filtering method. The results showed that the polycyclic lactone compound UK-78623 was first isolated from the fermentation of Streptomyces sp. FJS31-2, which had a significant inhibiting effect on Yersinia enterocolitica, and the diameter of antibacterial circle was 25.2 mm.
