摘要
【目的】筛选白芨试管球茎膨大培养基配方,为白芨的离体快繁和工业化生产提供参考依据。【方法】采用L9(34)正交试验和单因素完全随机试验设计方法,筛选适宜白芨试管球茎膨大的基本培养基、生长调节剂种类和浓度及蔗糖和活性炭浓度,确定白芨试管球茎膨大的最佳培养基配方。【结果】萘乙酸(NAA)和基本培养基对白芨试管球茎膨大的影响大于6-苄氨基嘌呤(6-BA),当NAA浓度为0.1 mg/L时,试管球茎膨大的各指标值均最大;6-BA对试管球茎膨大各指标值均无显著影响(P>0.05);以1/2MS为基本培养基更有利于白芨试管球茎生根和膨大。蔗糖是影响白芨试管球茎膨大的重要因素,随蔗糖浓度增加,试管球茎逐渐膨大且根生长量及植株长势渐好,当蔗糖浓度为20.0~40.0 g/L时试管球茎增殖效果较佳。培养基中添加0.5 g/L活性炭的白芨试管球茎鲜重增殖率最高,为33.41%。相关性分析结果表明,试管球茎生根率与鲜重增殖率呈正相关,与高度增殖率呈显著正相关(P<0.05);试管球茎萌芽率与鲜重增殖率、高度增殖率和直径增殖率均呈负相关,即生根对试管球茎膨大起促进作用,而芽分化对试管球茎膨大起抑制效应。【结论】1/2MS+0.1 mg/L NAA+1.0 mg/L 6-BA+20.0 g/L蔗糖+0.5 g/L活性炭可作为白芨试管球茎膨大的最佳培养基配方加以推广应用。
        【Objective】In order to provide reference for in vitro rapid propagation and industrial production of Bletilla striata,the medium formulation of bulbous enlargement was screened.【Method】The suitable basic medium,different plant growth regulators,concentrations of sucrose and activated carbon on B. striata bulbous enlargement were studied utilizing L9(34)orthogonal experiment and single-factor complete random experiment design methods to screen the best medium formulation for bulbous enlargement of B. striata in vitro.【Result】The results showed that the effects of nafusaku(NAA)and basal medium on bulbous enlargement was greater than that of 6-benzylaminopurine(6-BA). When NAA concentration was 0.1 mg/L,the average value of each index of bulb enlargement was the largest. 6-BA had no significant effect on each index of bulbous enlargement(P>0.05). 1/2 MS was beneficial to the rooting and bulbous enlargement of B.striata. Sucrose was an important factor affecting the bulbous enlargement of B. striata in vitro. With the increase of sucrose content,the bulb gradually expanded,the roots and plant growth were fine. The bulb proliferation of B. striata was fine when sucrose concentration was 20.0-40.0 g/L. The fresh weight proliferation rate was the highest(33.41%)with 0.5 g/L activated carbon adding into basal medium. Correlation analysis showed that the rooting rate was positively correlated with fresh weight gain rate,and was significantly positively correlated with height multiplication rate(P<0.05). Germination rate was negatively correlated with fresh weight gain rate,height multiplication rate and diameter multiplication rate.It indicated that rooting promoted bulbous enlargement and the bud differentiation inhibited bulb enlargement.【Conclusion】The suitable medium formulation for the bulbous enlargement of B. striata is 1/2 MS+0.1 mg/L NAA+1.0 mg/L6-BA+20.0 g/L sucrose+0.5 g/L activated carbon and can be promoted in production.
