摘要
目的了解雌激素受体(ER)-β基因型对壮族妇女绝经后其血清促卵泡激素、黄体生成素和雌孕激素的影响,为老年女性雌激素缺乏的疾病的提前防治提供个体化方案。方法对379例壮族(广西)45~50岁绝经妇女清晨空腹常规采静脉血5ml,使用PCR-RFLP方法检测并分析ER-β基因分布情况,用放射免疫试剂盒法测定各基因型样本血清的促卵泡激素、黄体生成素、雌二醇和孕激素。结果在对5个ER-β基因RsaI酶切基因(RR、Rr、rr、r、R)组的血清促卵泡激素、黄体生成素和雌二醇、孕酮水平比较中发现:RR基因组、Rr基因组、rr基因组、R基因组,4组间两两比较,差异无统计学意义(P>0.05);而r基因组与其它4组之间比较,差异有统计学意义(P<0.05),r基因组的血清雌二醇和孕酮水平明显低于其它4组(P <0.05),而r基因组的促卵泡激素、黄体生成素明显高于其它4组(P <0.01)。对5个ER-β基因AluI酶切基因(AA、Aa、aa、a、A)组的血清雌二醇、孕酮水平比较中发现,AA基因组、Aa基因组、a基因组、A基因组4组间相比较差异无统计学意义(P>0.05);aa基因组与其它4组之间比较差异有统计学意义(P <0.01),aa基因组的血清雌二醇和孕酮水平明显高于其它4组(P <0.05),而aa基因组的促卵泡激素、黄体生成素明显低于其它4组(P <0.01)。结论携带ER-βRsaI酶切的r等位基因绝经后妇女雌孕激素水平比较低,更易患绝经后雌激素缺乏性老年性疾病,应做好提前防治。而携带ER-βAluI酶切基因的aa等位基因型的绝经后妇女,其雌孕激素水平较高,不易患雌激素缺乏性疾病。
        Objective To understand the effects of estrogen receptor(ER)-βgenotype on serum follicle stimulating hormone,luteinizing hormone and estrogen and progesterone in Zhuang women after menopause,and to provide individualized treatment for early prevention and treatment of estrogen deficiency diseases in elderly women. Methods Each of 379 Guangxi Zhuang nationality postmenopausal women aged 45 to 50 years was routinely drawn 5 ml of venous blood in the morning,and the distribution of ER-βgene was detected and analyzed by PCR-RFLP.The follicle stimulating hormone,luteinizing hormone and estradiol and progesterone in serum samples of each genotype were determined by radioimmunoassay kit. Results Comparison of the serum follicle stimulating hormone,luteinizing hormone and estradiol,progesterone levels in the five ER-βrestriction enzyme RsaI genomes(RR,Rr,rr,r,R)indicated that pairwise comparison among RR genome,Rr genome,rr genome and R genome yielded no statistically significant difference(P >0.05).Comparison between r genome and the other four groups yielded significant difference(P <0.05),the serum estradiol and progesterone expressions in r genome were significantly lower than those of the other 4 genomes(P <0.05),while the follicle stimulating hormone and luteinizing hormone in the r genome were significantly higher than those of the other 4 genomes(P <0.01).Comparison of serum estradiol,and progesterone levels in the 5 ER-βgene restriction enzyme AluI genomes(AA,Aa,aa,a,A)indicated that pairwise comparison among AA genome,Aa genome,agenome and A genome yielded no statistically significant difference(P >0.05).Comparison between the aa genome and the other 4 genomes yielded statistically significant difference(P <0.01),the serum estradiol and progesterone levels in the aa genome were significantly higher than those of the other 4 genomes(P <0.05),while follicle stimulating hormone and luteinizing hormone in aa genome were significantly lower than those of the other 4 genomes(P <0.01). Conclusion The ER-βrestriction enzyme RsaI r allele carriers in postmenopausal women have a lower level of estrogen and progesterone,which is more prone to have senile postmenopausal estrogen deficiency diseases so that prevention and treatment should be done in advance.The ER-βrestriction enzyme AluI aa allele carriers in menopausal women have higher levels of estrogen and progesterone and are less prone to have estrogen deficiency disease.
