摘要
烟曲霉(Aspergillus fumigatus)是一种常见的通过空气传播的条件性致病丝状真菌,其生存环境受到扰动会释放大量的分生孢子(conidia),导致免疫能力低下的人患侵染性曲霉病(Invasive aspergillosis,IA)。增加对真菌新型靶向治疗途径的认知,可有效地预防和治疗疾病。然而,目前对丝状真菌中的自噬(Autophagy)机制尚未进行完全探索。自噬在丝状真菌中发挥重要作用,如饥饿期间参与营养物质的回收和再利用,促进分生孢子和菌丝觅食调节其正常的生长发育。自噬作为细胞器和细胞质成分等细胞内容物的一种新型降解途径,其激活可能与真菌快速的生长不相容,进一步探讨并通过药理学干预选择性诱导真菌自噬,可能会提供一个新的靶点。在真核生物中,c AMP信号通路参与多种调节过程,且已有研究证明这一信号通路对免疫抑制的IA啮齿类动物模型的毒力至关重要。c AMP途径的下游的主调控因子是蛋白激酶A(PKA),PKA是细胞中调节生长、代谢和增殖的重要信号分子之一。本实验选取PKA调节亚基缺失突变株(ΔPka R)的烟曲霉,初步探讨烟曲霉中PKA与自噬的关系,为揭示该基因在烟曲霉的致病中的作用提供一定的理论基础。
        Aspergillus fumigatus is a common airborne condition-causing filamentous fungus that perturbs its living environment and releases large amounts of conidia, leading to people with low immunity. Invasive aspergillosis(IA). Increased cognition of new targeted therapeutic approaches for fungi can effectively prevent and treat diseases. However, the current mechanism for autophagy in filamentous fungi has not been fully explored. Autophagy plays an important role in filamentous fungi, such as participating in the recovery and reuse of nutrients during starvation, promoting conidial and mycelial foraging to regulate its normal growth and development. Autophagy as a cellular content, including a novel degradation pathway for organelles and cytoplasmic components, its activation may be incompatible with rapid growth of fungi, and further exploration and selective induction of fungal autophagy through pharmacological intervention may provide a new target.In eukaryotes, the c AMP signaling pathway is involved in a variety of regulatory processes, and studies have demonstrated that this signaling pathway is critical for the virulence of immunosuppressed IA rodent models. The main downstream regulator of the c AMP pathway is PKA, one of the important signaling molecules that regulate growth, metabolism, and proliferation in cells. In this experiment, PKA was used to select the Aspergillus fumigatus mutant subunit deletion mutant(ΔPka R), and the relationship between PKA and autophagy in Aspergillus fumigatus was preliminarily investigated, which provided a theoretical basis for revealing the role of this gene in the pathogenesis of Aspergillus fumigatus.
