温胆汤对肥胖大鼠血清瘦素及下丘脑STAT3和SOCS3表达的影响

摘要
目的探讨温胆汤治疗肥胖的可能作用机制。方法采用高脂饮食诱导肥胖大鼠模型,将造模成功大鼠随机分成模型组和温胆汤组,每组8只,另将8只健康大鼠设为正常组。造模成功后各组大鼠均采用基础饲料喂养,同时温胆汤组大鼠给予温胆汤15 g/(kg·d)灌胃,模型组和正常组均用等量蒸馏水灌胃,共持续6周。比较各组大鼠体重、Lee's指数、肥胖率,检测血清瘦素、血脂[包括总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)和高密度脂蛋白胆固醇(HDL-C)]及下丘脑组织信号转导和转录激活因子3 (STAT3)、细胞因子信号转导抑制因子3 (SOCS3) mRNA及蛋白表达。结果与正常组比较,模型组大鼠体重、血脂水平(除HDL-C外)、下丘脑STAT3 mRNA和蛋白表达升高,SOCS3 mRNA和蛋白表达降低(P <0. 01)。与模型组比较,温胆汤组Lee's指数、体重、血脂水平(除HDL-C外)、下丘脑STAT3 mRNA和蛋白表达明显降低,SOCS3 mRNA和蛋白表达则显著升高(P <0. 01)。模型组肥胖率为26. 36%,温胆汤组为1. 06%。结论温胆汤具有良好的降脂作用,可能通过调节JAK2/STAT3信号通路而调控机体瘦素水平,从而纠正机体代谢异常。
        Objective To investigate the possible mechanism of Wendan Decoction(温胆汤) in the treatment of obesity. Methods A rat model of obesity induced by high-fat diet was randomly divided into model group and Wendan Decoction group,with 8 rats in each group,and 8 healthy rats were set as normal group. After successful modeling,the rats in each group were fed with basal diet. The rats in Wendan Decoction group were given 15 g/( kg·d) of Wendan Decoction. The model group and the normal group were treated with the same amount of distilled water,for 6 weeks. The body weight,Lee's index,and obesity rate of each group were compared. Serum leptin and blood lipids including total cholesterol( TC),triglyceride( TG),low density lipoprotein cholesterol( LDL-C) and high-density lipoprotein cholesterol( HDL-C)) and hypothalamic signal transducers and activators of transcription 3( STAT3),suppressor of cytokine signaling 3( SOCS3) mRNA and protein expression were measured. Results Compared with the normal group,the body weight and blood lipid levels( except HDL-C),hypothalamic STAT3 mRNA and protein expression,and SOCS3 mRNA and protein expression were decreased in the model group( P < 0. 01). Compared with the model group,the Lee's index,body weight,blood lipid level( except HDL-C),hypothalamic STAT3 mRNA and protein expression were significantly decreased,and SOCS3 mRNA and protein expression were significantly increased( P < 0. 01). The obesity rate of the model group was 26. 36%,and that of the Wendan Decoction group was 1. 06%. Conclusion Wendan Decoction has a good lipid-lowering effect,which may regulate the body's Leptin level by regulating JAK2/STAT3 signaling pathway,thus correcting the body's metabolic abnormalities.
