摘要
目的探讨丹参酮的抗辐射损伤作用。方法建立紫外线损伤小鼠模型。取30只小鼠,随机分为6组,每组5只,即Ⅰ组(正常对照组,生理盐水),Ⅱ组(模型对照组),Ⅲ组(模型对照组,维生素C 5 g/L),Ⅳ~Ⅵ组为3个剂量丹参酮组(2. 5,5. 0,10. 0 g/L)。适应环境1周后,每日上午给予Ⅰ组和Ⅱ组0. 5 m L生理盐水灌胃,Ⅲ组0. 5 m L维生素C注射液灌胃,Ⅳ~Ⅵ组为0. 5 m L的各质量浓度梯度的丹参酮注射液灌胃;Ⅱ~Ⅵ组实验动物下午均接受10 min的紫外线照射,14 d后,取小鼠眼眶血并处死,采用比色法测定小鼠血清中谷胱甘肽还原酶(GR)、谷胱甘肽(GSH)、总抗氧化能力(T-AOC)、丙二醛(MDA)的活力或含量。结果与Ⅰ组相比,Ⅱ组小鼠的血清GR活力、T-AOC能力、GSH含量均明显下降,MDA含量明显上升(P <0. 05);与Ⅱ组相比,Ⅲ~Ⅵ组小鼠的血清GR活力、T-AOC能力、GSH含量均明显上升,MDA含量明显下降(P <0. 05);与Ⅳ~Ⅵ组相比,Ⅲ组小鼠的血清GR活力、T-AOC能力、GSH含量、MDA含量无显著差异(P> 0. 05)。结论丹参酮能清除氧自由基,降低紫外辐射对机体的损伤。
        Objective To investigate the protective effect of tanshinone on radiation injury. Methods The mice models of ultraviolet injury were established. Totally 30 mice were randomly divided into 6 groups with 5 mice in each group: Group Ⅰ( normal control group,normal saline),group Ⅱ( model control group),group Ⅲ( model control group,5 g/L vitamin C),groups Ⅳ-Ⅵ( 2. 5,5. 0,10. 0 g/L tanshinone). One week after acclimatization to the environment,the mice in the group Ⅰ and groups Ⅱ received intragastric administration of 0. 5 m L normal saline daily in the morning,the mice in the group Ⅲ received intragastric administration of 0. 5 m L Vitamin C Injection,and the mice in the groups Ⅳ-Ⅵ received intragastric administration of 0. 5 m L Tanshinone Injection with different dosages. The mice in the groups Ⅱ-Ⅵreceived ultraviolet light for 10 min in the afternoon. After 14 d of treatment,the orbital blood of mice was taken and the mice were sacrificed,the activity or content of glutathione reductase( GR),glutathione( GSH),total antioxidant capacity( T-AOC) and malondialdehyde( MDA) in serum of mice were determined by colorimetry. Results Compared with group Ⅰ,the serum GR activity,T-AOC ability and GSH content of the mice in group Ⅱ decreased significantly,while the MDA content increased significantly( P < 0. 05). Compared with group Ⅱ,the serum GR activity,T-AOC ability and GSH content of the mice in groups Ⅲ-Ⅵ increased significantly,while the MDA content decreased significantly( P < 0. 05). There were no significant differences in serum GR activity,T-AOC ability,GSH content and MDA content among the groups Ⅳ-Ⅵ and group Ⅲ( P > 0. 05). Conclusion Tanshinone can eliminate oxygen free radicals and reduce the injury of ultraviolet radiation to the body.
