摘要
目的建立高效液相色谱法测定山柰药材中槲皮素和山柰素的含量。方法采用ODS2 C18(5μm,4.6 mm×150 mm)色谱柱,以甲醇-0.4%磷酸(47∶53)为流动相,流速1 m L/min,柱温30℃,检测波长为367 nm,进样量为10μL。结果槲皮素在0.016 5~1.65μg范围内呈现良好的线性关系(r=0.999 7),平均回收率为96.8%,RSD=2.02%;山柰素在0.014 6~1.46μg范围内呈现良好的线性关系(r=0.999 5),平均回收率为97.3%,RSD=1.77%。结论该方法操作简单、结果准确、重复性较好,可用于山柰药材中槲皮素和山柰素的含量测定。
Objective To establish an HPLC method for the simultaneous determination of quercetin and kaempferol in Kaempferia galanga L.. Methods ODS2 C18(5 μm, 4.6 mm×150 mm) was used as chromatographic column; methanol-0.4% phosphate(47:53) was the mobile phase; the flow rate was 1 m L/min; column temperature was 30 ℃; the detection wavelength was 367 nm; the injection volume was 10 μL. Results Quercetin showed good linear relationship in the range of 0.016 5–1.65 μg(r=0.999 7). The average recovery rate was 96.8%, RSD=2.02%; kaempferol showed good linear relationship in the range of 0.014 6–1.46 μg(r=0.999 5). The average recovery rate was 97.3%, RSD=1.77%. Conclusion The method is simple, accurate, and with good reproducibility, which can be used for content determination of quercetin and kaempferol in Kaempferia galanga L..
