摘要
目的分析反式激活活性蛋白(C-myc)与LIN28B mRNA、蛋白在胃癌组织和相应癌旁正常组织中的表达及其与胃癌发生发展的相关性。方法通过Western blot法对60例胃癌组织和相应癌旁正常组织中C-myc、LIN28B蛋白表达水平进行检测,实时荧光定量PCR法测定C-myc、LIN28B mRNA表达水平,统计分析C-myc、LIN28B蛋白表达与胃癌临床病理参数的关系。结果 C-myc、LIN28B蛋白在胃癌组织中的表达水平较癌旁正常组织均明显升高(P<0. 01); C-myc、LIN28B mRNA在胃癌组织中的表达水平较癌旁正常组织均明显升高(P <0. 01)。C-myc蛋白表达与胃癌TNM分期、浸润深度及淋巴结转移均密切相关(P <0. 05); LIN28B蛋白表达与胃癌TNM分期、分化程度、浸润深度及淋巴结转移均密切相关(P <0. 05)。结论 C-myc、LIN28B蛋白及其mRNA均高表达于胃癌组织,二者与胃癌TNM分期、浸润深度及淋巴结转移均密切相关,可能共同参与胃癌的病理过程。
        Objective To analyze the expression of trans activator protein( C-myc) and LIN28 B mRNA and protein in gastric cancer tissues and adjacent normal tissues and correlation with the occurrence and development of gastric cancer. Methods The Western blot method was used to detect the level of C-myc and LIN28 B protein expression in 60 cases of gastric cancer tissues and adjacent normal tissues,the expression levels of C-myc and LIN28 B mRNA were detected by the real-time fluorescence quantitative PCR method. The correlation between C-myc,LIN28 B protein expression and clinicopathological parameters of gastric cancer was statistically analyzed. Results The expression levels of C-myc and LIN28 B protein in gastric cancer tissues were significantly higher than those in adjacent normal tissues( P < 0. 01). The expression levels of C-myc and LIN28 B mRNA in gastric cancer tissues were significantly higher than those in adjacent normal tissues( P < 0. 01). C-myc protein expression was closely correlated with TNM staging,depth of invasion and lymph node metastasis of gastric cancer( P < 0. 05). LIN28 B protein expression was closely correlated with TNM staging,differentiation,invasion depth and lymph node metastasis of gastric cancer( P < 0. 05).Conclusion C-myc,LIN28 B protein and mRNA are all highly expressed in gastric cancer tissues,which are closely correlated with TNM stage,depth of invasion and lymph node metastasis,and may participate in the pathological process of gastric cancer.
