摘要
目的探究内皮素-1(ET-1)对正常犬心脏右侧星状神经节(RSG)功能及神经活性的影响。方法 12只成年犬随机均分为两组:ET-1组(n=6,给予RSG局部微注射0.15μmol/ml ET-1 0.1ml)和对照组(n=6,给予RSG局部微注射等量0.9%生理盐水)。两组动物均于麻醉后行右侧开胸术,暴露RSG。分别在基础状态和注射ET-1或生理盐水30min后检测RSG在高频电刺激(HFS,频率:20 Hz,脉宽:0.1 ms)下,最大窦性心率(sinusrate,SR)的变化情况,此即RSG功能。在相同时间点记录RSG的神经活性。结果在基础状态下,HFS刺激RSG引起的最大SR改变和RSG神经活性在ET-1组和对照组中无差异(P>0.05)。但在RSG局部注射ET-1 30min后,与对照组相比,RSG的功能显著增强[20V:(46.0±4.2)%vs(37.7±5.6)%,P<0.05],HFS刺激RSG引起的最大SR改变也明显增强[频率:(95±11)次/分vs(33±7)次/分;振幅:(0.20±0.04)mV vs(0.10±0.04)mV,P均<0.05]。结论 ET-1可显著增强RSG的功能及神经活性。
Objective To investigate the effects of endothelin-1(ET-1)on right stellate ganglion(RSG)function and neural activity in healthy normal canines. Methods Twelve anesthetized dogs were randomly divided into ET-1 group(n=6,0.1 ml of 0.15μmol/ml ET-1 was microinjected into RSG)and control group(n=6,equivalent saline was microinjected into RSG).The changes of sinus rate(SR)in high frequency electrical stimulation(HFS,parameters:20 Hz in frequency,0.1 ms in pulse width)on RSG(RSG function)and its neural activity were determined at baseline and 30 min after microinjection. Results At baseline,significant differences were not observed on the function and neural activity of RSG between the ET-1 group and the control group.However,30 min after ET-1 microinjection,the function[20 V:(46.0±4.2)% vs(37.7±5.6)%,P<0.05]and neural activity of RSG[frequency:(95±11)impulses/min vs(33±7)impulses/min;amplitude:(0.20±0.04)mV vs(0.10±0.04)mV,P<0.05 for both]were significantly increased in the ET-1 group compared to that in the control group. Conclusion ET-1 could significantly enhance the function and neural activity of RSG.
