摘要
目的探讨坦索罗辛通过miRNA改善大鼠前列腺炎的作用。方法利用角叉菜胶溶液诱导大鼠前列腺病变,建立前列腺炎小鼠模型后,HE染色及天狼星红染色观察前列腺组织病理形态。应用Real-time PCR技术检测坦索罗辛对大鼠炎症模型及大鼠前列腺上皮细胞炎症模型中miR-141表达影响;应用Western blot检测坦索罗辛对大鼠前列腺组织及前列腺上皮细胞中TGF-β_1和α-SMA蛋白表达影响;应用试剂盒检测坦索罗辛对前列腺组织及前列腺上皮细胞中炎症因子TNF-α表达水平影响。结果坦索罗辛能抑制miR-141在大鼠前列腺组织中的下调,同时抑制前列腺的弥漫性炎症、胶原聚集、TNF-α,TGF-β_1及α-SMA的表达;给予人前列腺细胞炎症模型坦索罗辛治疗并敲减miR-141,可逆转坦索罗辛的治疗作用。结论坦索罗辛能通过调控miR-141表达改善前列腺炎,为坦索罗辛临床应用提供理论基础。
