补阳还五汤、少腹逐瘀汤、丹参饮对寒凝血瘀模型大鼠血小板形态与黏附的影响

摘要
目的从血小板黏附以及纤溶功能分析补阳还五汤、少腹逐瘀汤、丹参饮对寒凝血瘀的治疗的机制。方法选取80只大鼠,按体质量随机分成空白组和寒凝血瘀模型组,补阳还五汤、少腹逐瘀汤和丹参饮高低剂量组,10只/组。大鼠复制寒凝血瘀模型,连续造模15 d,每日灌胃给药,末次颈总动脉取血,测定血小板相关的各项指标。结果与空白组比较,模型组血小板体积分布宽度(PDW)显著升高(P <0.01),纤维连接蛋白(FN)含量显著升高(P <0.05)。与模型组比较,各给药组PDW显著降低(P <0.05),血小板平均体积(MPV)显著降低(P <0.05);丹参饮低剂量组血管性血友病因子(vWF)、FN含量显著降低(P <0.05),其余无显著变化。结论补阳还五汤、少腹逐瘀汤、丹参饮可能是通过降低PDW、MPV,由此降低血小板活性,从而起到了抗栓的作用,其中丹参饮降低血浆内v WF和FN水平,保护内皮细胞,可能是治疗寒凝血瘀证的机制之一。
        Objective According to platelet adhesion and fibrinolysis function, the therapeutic mechanism of Buyang Huanwu decoction, Shaofu Zhuyu decoction and Danshen decoction on cold coagulation and blood stasis was analyzed. Methods 80 rats were randomly divided into blank group and cold coagulation and blood stasis model group according to their body weight, and the low and high dose groups of Buyang Huanwu decoction, Shaofu Zhuyu decoction and Danshen decoction group, each group contained 10 rats. The rat model of cold coagulation and blood stasis was established, and the model was established for 15 d continuously, intragastric administration was given every day, and the blood samples were taken from the common carotid artery. Results Compared with the blank group, the PDW of the model group was significantly increased(P < 0.01), the content of FN was significantly increased(P < 0.05). Compared with the model group, each group PDW significantly decreased(P < 0.05), MPV were significantly decreased(P < 0.05). Danshen decoction low dose group vWF, FN was significantly decreased(P < 0.05), while no significant change. Conclusion Buyang Huanwu decoction, Shaofu Zhuyu decoction and Danshen decoction may reduce PDW and MPV, which can reduce the activity of platelet, and thus play a role in anti thrombosis. Danshen decoction can decrease the levels of vWF and FN in the plasma and protect the endothelial cells, which may be one of the mechanisms of the treatment of cold coagulation and blood stasis syndrome.
