补中益气汤对Lewis肺癌小鼠EGFR、NF-κB、ICAM-1表达及JAK1、STAT1蛋白磷酸化的影响

摘要
目的:探讨补中益气汤对Lewis肺癌小鼠表皮生长因子(epidermal growth factor receptor,EGFR),核转录因子κB(nuclear factor kappa beta,NF-κB),细胞间黏附分子-1(intercellular cell adhesion molecule-1,ICAM-1)表达及蛋白质酪氨酸激酶-1(janus kinase-1,JAK1),转录活化蛋白1(signal transducer and activator of transcription 1,STAT1)蛋白磷酸化的影响。方法:选取SPF级小鼠60只,随机分为空白组、模型组、西药组、补中益气汤高剂量组、补中益气汤中剂量组、补中益气汤低剂量组,每组10只。除空白组外均将Lewis肺癌细胞无菌接种于小鼠的右腋皮下,建立Lewis肺癌模型。造模成功后,模型组以无菌生理盐水0.2 m L灌胃,西药组小鼠给予吉非替尼50 mg·kg~(-1),每次0.2 m L;补中益气汤高、中、低剂量组以补中益气汤20 g·kg~(-1)、10 g·kg~(-1)、5 g·kg~(-1)进行灌胃,每次0.2 m L,灌胃均为每天1次。各组用药2周。观察并比较各组小鼠的瘤质量、瘤指数、NF-κB、ICAM-1、EGFR、JAK1、STAT1水平。结果:与模型组相比,西药组、补中益气汤高剂量组、补中益气汤中剂量组小鼠的瘤质量和瘤指数,NF-κB、ICAM-1、EGFR及JAK1、STAT1水平较低,差异具有统计学意义(P<0.05);与西药组相比,补中益气汤高剂量组、补中益气汤中剂量组小鼠的瘤质量和瘤指数,NF-κB、ICAM-1、EGFR、JAK1、STAT1水平较低,差异具有统计学意义(P<0.05);补中益气汤高剂量组、补中益气汤中剂量组小鼠的瘤质量和瘤指数,NF-κB、ICAM-1、EGFR、JAK1、STAT1水平相比,差异无统计学意义(P>0.05)。结论:补中益气汤能够降低Lewis肺癌小鼠瘤质量和瘤指数,降低EGFR,NF-κB、ICAM-1、JAK1、STAT1水平,对临床具有一定的参考意义。
        Objective:To investigate the effects of Buzhong Yiqi Decoction on epidermal growth factor receptor( EGFR),nuclear factor kappa beta( NF-κB),cell adhesion molecule-1( ICAM-1) and phosphorylation of protein tyrosine kinase-1( JAK1) and STAT1 protein( STAT1) in Lewis lung cancer mice. Methods:60 SPF mice were randomly divided into blank group,model group,western medicine group,Buzhong Yiqi Decoction high dose group,Buzhong Yiqi Decoction group,and Buzhong Yiqi Decoction low dose group,with 10 rats in each group. Except for the blank group,Lewis lung cancer cells were established aseptically inoculated into the right iliac crest of mice and a lung cancer tumor model was established. After the model was successfully established,the model group was given 0. 2 m L of sterile normal saline. The western medicine group mice were treated with gefitinib( CTX) 50 mg·kg~(-1),0. 2 m L each time;Buzhong Yiqi Decoction high,middle,and The low-dose group was given intragastric administration of Buzhong Yiqi Decoction at 20 g·kg~(-1),10 g·kg~(-1),5 g·kg~(-1) each time,0. 2 m L each time,and the intragastric administration was performed once daily. The two groups used medication for 2 weeks. The tumor mass,tumor index,NF-κB,ICAM-1,EGFR and JAK1,STAT1 levels were observed and compared in each group of mice. Results:Compared with the model group,the tumor quality and tumor index levels of the mice in the Western medicine group,the high-dose group of Buzhong Yiqi Decoction,and the middle-dose group of Buzhong Yiqi Decoction,NF-κB,ICAM-1,EGFR,and JAK1 The level of STAT1 was lower,and the difference was statistically significant( P < 0. 05). Compared with the western medicine group,the tumor quality and tumor index level of the high-dose Buzhong Yiqi Decoction group and the middle-dose group of Buzhong Yiqi Decoction. The levels of NF-κB,ICAM-1,EGFR and JAK1,STAT1 were lower,the difference was statistically significant( P < 0. 05); the tumor quality of mice in high dose of Buzhong Yiqi Decoction and middle dose of Buzhong Yiqi Decoction was significant The tumor index level,NF-κB,ICAM-1,EGFR,JAK1,and STAT1 levels had no significant difference( P > 0. 05). Conclusion:Buzhong Yiqi Decoction can reduce the tumor quality and tumor index of Lewis lung cancer mice,and decrease the levels of EGFR,NF-κB,ICAM-1,JAK1 and STAT1.
