摘要
为了探讨外源H_2O_2对盐胁迫下黄瓜种子萌发抑制作用的缓解机制,以黄瓜品种‘津瑞100’种子为材料,采用培养皿滤纸发芽法,测定了不同浓度H_2O_2浸种处理对150 mmol·L~(-1) NaCl胁迫下黄瓜种子萌发及相关生理指标的影响。结果表明,在150 mmol·L~(-1) NaCl胁迫下,黄瓜种子的萌发受明显抑制, MDA含量显著增加,抗氧化酶(SOD、POD、CAT、APX)的活性降低; ABA含量显著增加,而GA含量显著降低。适宜浓度的H_2O_2浸种处理能不同程度提高盐胁迫下黄瓜种子的萌发率和抗氧化酶(SOD、POD、CAT、APX)的活性;降低MDA和ABA含量,增加GA含量,从而缓解盐胁迫对黄瓜种子萌发的抑制作用。其中以0.3%H_2O_2浸种处理效果最好。高浓度H_2O_2处理对盐胁迫的缓解作用逐渐减弱甚至加重其毒害效应。说明适宜浓度的H_2O_2可以通过介导抗氧化酶系统、ABA和GA缓解盐胁迫对黄瓜种子萌发的抑制作用。
        In order to explore the ameliorating mechanism of hydrogen peroxide(H_2O_2) on inhibition of seed germination of cucumber caused by NaCl, the effects of different H_2O_2 concentration on seed germination and physiological characteristics of cucumber(‘Jinrui No. 100’) were studied under 150 mmol·L~(-1) NaCl stress. The results showed that the seed germination of cucumber was inhibited significantly by 150 mmol·L~(-1) NaCl stress. MDA content increased significantly, the activities of SOD, POD, CAT and APX were inhibited obviously. On the other hand, ABA content increased significantly, GA content decreased obviously. Suitable concentration of H_2O_2 could alleviate the inhibition effects of NaCl. Compared to 150 mmol·L~(-1) NaCl stress, the seed germination percent, the activities of SOD, POD, CAT and APX, and GA content could be improved significantly by pretreated with 0.3% H_2O_2, while ABA content significantly decreased. Moreover, we found that the mitigation effect of NaCl stress was gradually weakened with the increase of H_2O_2 concentration, and 1% H_2O_2 would aggravate the toxicity of NaCl stress. In this study, we found that the appropriate concentration of H_2O_2 could alleviate the inhibitory effects of NaCl stress on germination mainly by regulating antioxidant systems, ABA and GA interaction in cucumber.
