摘要
目的:建立简化样品处理快速测定酸枣仁中多种化学成分含量的方法。方法:比较酸枣仁样品的不同前处理方法,包括简单超声提取法、直接加热回流法和脱脂后加热回流提取法(药典方法)下UPLC-UV-ELSD液相检测黄酮类和皂苷类等4种成分的峰面积。结果:通过比较,药典方法在酸枣仁脱脂后回流4h反而导致成分含量降低,加热回流30 min提取率最高,以此对比,超声90 min对酸枣仁中皂苷类和黄酮类成分的提取效果相当,提取溶剂为70%乙醇,物料比为1∶30。结论:本文建立的提取方法可以获得提取充分的效果,且快速和易于操作,为控制酸枣仁中黄酮类成分和皂苷类成分含量的快速测定提供了可靠的方法。
        Objective:To establish a simple and rapid method for the determination of various chemical components.Methods:Through UPLC-UV-ELSD liquid phase method spinosin,6-feruloyl spinosin,jujuboside A and jujuboside B 4 kinds of ingredients such as peak area,different pretreatment methods were compared for the samples of spina date seed,including simple ultrasonic extraction,direct heating reflux and degreased and then heated reflux extraction.Results:By comparison,the reflux of 4 hours after the degreasing of Semen Ziziphi Spinosae resulted in the decrease of the content of the ingredients in Semen Ziziphi Spinosae.Heating reflux for 30 min was the best extraction method.By contrast,The effect of ultrasound at 90 min was comparable to that of reflux at 30 min.Therefore,the ultrasonic extraction method was recommended for 90 min.In ultrasonic extraction,the final optimum extraction condition were 70% ethanol as the extracting solvent,1∶30 as the material ratio.Conclusion:The method established in this paper can obtain sufficient extraction effect,and is fast and easy to operate.It provides a reliable method for rapid determination of flavonoids and saponins in Semen Ziziphi Spinosae.
