摘要
以澳洲坚果品种‘H2’幼苗的幼嫩茎段为外植体材料,分别采用WPM、MS、1/2MS培养基,分析添加不同植物生长调节剂及其配比对腋芽启动、增殖及生根的影响。结果表明:适合茎段腋芽启动的培养基为WPM+6-BA 2.0 mg/L+IBA 1.0 mg/L+AC 200 mg/L,诱导系数为3.51,腋芽萌发后生长健壮;最佳增殖培养基为MS+6-BA 2.0 mg/L+KT 0.5 mg/L,增殖系数达3.49;组培苗诱导生根培养基为1/2MS+6-BA 0.2 mg/L+IBA 0.5 mg/L+ABA 0.5 mg/L+AC 200 mg/L,生根率为42.2%。
The shoot segments of macadamia nut cultivar 'H2' were used as explant materials, and WPM, MS and 1/2 MS were used as the basic medium to study the effects of different exogenous hormones and their ratios on axillary bud initiation, proliferation and rooting of tissue cultured seedlings. The results show that suitable medium for axillary bud initiation is WPM + 6-BA 2.0 mg/L + IBA 1.0 mg/L + AC 200 mg/L, and the induction coefficient is 3.51. Meanwhile, the sprouts grow robustly after axillary buds germination. The optimal proliferation medium for axillary buds is MS + 6-BA 2.0 mg/L + KT 0.5 mg/L, and the proliferation coefficient reaches 3.49.The optimum rooting formula selected in this study is 1/2 MS + 6-BA 0.2 mg/L + IBA 0.5 mg/L + ABA 0.5 mg/L +AC 200 mg/L, the rooting rate is 42.2%.
