摘要
目的:研究归芪多糖对衰老的WI-38细胞的细胞周期及p53、p16蛋白表达的影响。方法:采用MTT法,研究不同浓度归芪多糖对WI-38细胞活力的影响,筛选出归芪多糖最佳作用浓度;采用流式细胞术分析细胞周期;Western blot法分析归芪多糖对p53、p16蛋白表达的影响。结果:0.016/mg·m L归芪多糖可明显促进细胞增殖;与衰老组细胞相比,归芪多糖可明显促进衰老细胞进入分裂期(P<0.01),并可显著降低衰老模型组细胞p53和p16蛋白的表达。结论:归芪多糖可延缓WI-38细胞的衰老,其作用机制与调控细胞周期,抑制p53-p21和p16-pRb信号通路有关。
        Objective: To investigate the effect of Guiqi polysaccharide( GQP) on the cell cycle and the protein expression of p53 and p16 in senescence cells. Methods: MTT assay was used to observe the effect of different concentrations of GQP on the proliferation of WI-38 cells and screen the best concentration of GQP to promote the growth of WI-38 cells. Cell cycle was analyzed by flow cytometry,and Western blot analysis was performed to evaluate the protein expression of p53 and p16 in senescence cells. Results: 0. 016 mg / m L GQP was the best concentration of promoting cell proliferation. Compared with H2O2 model group,GQP could promote the senescence cells from G1-phase to S-phase and reduce the protein expression of p53 and p16,and the difference was significant( P < 0. 01). Conclusion: GQP has an protective effect on senescence cells. The anti-aging effect of GQP may be involved in regulating the cell cycle and weakening two signal pathways( p53-p21 and p16-pRb).
