摘要
AT模序结合因子1(AT-motif binding factor 1,ATBF1)又名为锌指同源框3(zincnger homeobox 3,ZFHX3),通过与启动子中的AT富含原件结合调控基因转录,是目前发现的相对分子质量最大的转录调节因子。近年来,越来越多的研究表明,ATBF1基因是一个重要的候选抑癌基因。ATBF 1基因在前列腺癌、胃癌、乳腺癌、宫颈癌和结直肠癌等多种恶性肿瘤中频繁发生基因缺失和突变、表达下调和蛋白定位异位等,通过多种信号途径参与细胞生命活动,影响细胞增殖、分化和凋亡,调控肿瘤的发生和发展。本文依次从DNA水平、mRNA水平、蛋白水平和分子机制4个方面对ATBF1基因在肿瘤中的研究进展进行综述,以期更全面综合地了解ATBF 1基因的抑癌机制及其对肿瘤诊断治疗的临床意义。
        AT-motif binding factor 1(ATBF1), also named as zinc finger homeobox 3(ZFHX3), is reported as a transcriptional regulator with the highest molecular weight. ATBF1 regulates mRNA transcription by binding to AT-rich motif of gene promoter. In recent years, more and more studies have shown that ATBF1 gene is an important candidate tumor suppressor gene. Frequent mutations and loss of heterozygosity(LOH) are found in ATBF1 gene in multiple tumors, including prostate cancer, gastric cancer, breast cancer, cervical cancer and colorectal cancer. The gene expression and protein localization of ATBF1 are associated with the clinical diagnosis and treatment of tumors. Moreover, ATBF1 plays an essential role in multiple signaling pathways to regulate cell proliferation, differentiation and apoptosis. This review summarizes the research progress of ATBF 1 gene in malignant tumors from four aspects: DNA, mRNA, protein and molecular mechanism, in order to comprehesively understand the anti-cancer mechanism of ATBF1 and its clinical significance in diagnosis and treatment of tumors.
