摘要
通过形态观察、生理生化试验、pH梯度生长试验以及拮抗性试验,对一株具有较强纤溶酶活力的放线菌701展开研究。初步鉴定为中度嗜酸放线菌,最优固体培养基为GYM固体培养基,最佳培养时间为5 d,最佳生长pH为5.5。最优发酵培养基成分为大豆粉2%,蛋白胨0.2%,葡萄糖2%,淀粉0.5%,酵母膏0.52%,CaCl_20.4%,K_2HPO_40.05%,MgSO_4·7H_2O 0.05%,CaCO_30.2%,最佳pH为7.8,最佳培养时间为5 d。拮抗性试验表明:该菌株本身对细菌、酵母及黑曲霉没有抑制作用,其发酵液除对大肠杆菌有抑制作用外,对其他细菌均无抑菌作用,相对最大抑菌环直径为第5天发酵液所得,抑菌环直径为36.2 mm。
Actinobacteria 701, which had high fibrinolytic enzyme activity, was identified as a Acidophilic actinomycete based on morphology, physiological biochemical test, pH range for growth and antagonistic test. The optimal solid medium was GYM medium(pH 5.5). The optimal fermentation media was composed of soybean flour 2%, peptone 0.2%, glucose 2%, starch 0.5%, yeast extract 0.5%, CaCl_(2 )0.4%, K_2HPO_(4 )0.05%, MgSO_4·7 H_2O 0.05%, CaCO_3 0.2%, pH 7.8. The optimal culture time is 5 days. The antagonistc test indicated that Actinomycete 701 has no antagonistic activity, while its fermentation broth showed antagonistic activity towards Escherichia coli. The diameter of the bacteriostatic annulus was 36.2 mm.
