摘要
目的:探索新扶正除疫颗粒剂对腺病毒感染小鼠肺损伤的防治效果。方法:取100只雌性Balb/c小鼠随机分为5组:病毒模型组,利巴韦林组,新扶正除疫颗粒低、中、高剂量组(简称低、中、高剂量组),5组小鼠建立腺病毒感染模型,并观察给药前后小鼠的一般状态、药物死亡保护作用,制作病理切片进行形态学观察、计算肺组织病毒滴度,腹主动脉采血后制备血清,ELISA法检测肿瘤坏死因子α(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)的表达水平。结果:与病毒模型组比较,新扶正除疫颗粒各剂量组小鼠一般状态均有不同程度的改善,死亡率显著降低(P<0.05,P<0.01)。病毒模型组小鼠肺组织纤维化,肺泡壁和间质增厚,肺泡内充满炎性渗出物等病理变化明显,新扶正除疫颗粒剂各剂量组炎性渗出物减少,病理变化明显减轻。与病毒模型组比较,新扶正除疫颗粒高、中剂量组小鼠肺组织内病毒滴度显著降低(P<0.05,P<0.01),与利巴韦林组比较差异无统计学意义,血清TNF-α、IL-1β、IL-6含量显著下降(P<0.05,P<0.01),IL-10表达显著增加(P<0.01)。结论:新扶正除疫颗粒剂对腺病毒感染导致肺损伤的小鼠具有明显保护作用。
        Objective: To explore the effects of New Fuzheng Chuyi Granules on lung injury in mice infected with adenovirus. Methods: One hundred female Balb/c mice were randomly divided into 5 groups: adenovirus model group, ribavirin control group, high, middle and low dosage groups of New Fuzheng Chuyi Granules. Five groups of mice developed adenovirus infection models, and the general state of the mice before and after administration and the protective effect of drug death were observed. Pathological sections were made for morphological observation, and the virus titer of the lung tissues was calculated. The serum was prepared from abdominal aorta blood. The levels of TNF-α, IL-1β, IL-6 and IL-10 were detected by ELISA. Results: Compared with the adenovirus model group, the general state of the mice in every group with New Fuzheng Chuyi Granules had improvement in varying degrees, and the death rates were also significantly lower(P<0.05, P<0.01). In the adenovirus model group, the pulmonary fibrosis, alveolar wall and interstitial thickening, alveolar inflammatory exudate and other pathological changes were obvious. After the New Fuzheng Chuyi Granules intervention, the inflammatory exudation decreased, and the pathological changes were significantly reduced. Compared with the model group, there was a significant decrease about virus titer in lung tissue of high and middle dosage groups New Fuzheng Chuyi Granules(P<0.05, P<0.01), while contrasted with the ribavirin group, the result was not significant. Compared with the model group, the contents of cytokines TNF-α, IL-1β, IL-6 in the high and middle dosage groups New Fuzheng Chuyi Granules were significantly decreased(P<0.05, P<0.01), and the IL-10 was significantly increased(P<0.01). Conclusion: The New Fuzheng Chuyi Granules has obvious protective effect on mice with lung injury caused by adenovirus infection.
