摘要
目的分析针刺治疗对于脑卒中疾病的痉挛状态中SD大鼠大脑皮质中多巴胺受体及其亚型的含量与表达的影响,探讨针灸缓解脑卒中痉挛状态(SCA)的作用机制。方法将32只SD大鼠随机分为针刺组(模型+常规手法针刺阳陵泉、曲池)、假手术组、模型组、空白组,每组8只。运用大脑中动脉线栓法制作SCA模型,行为学评分确认模型成功后开始针刺治疗,连续3 d。酶联免疫吸附实验法(ELISA)检测皮质多巴胺、多巴胺D1受体信使核糖核酸、多巴胺D2受体信使核糖核酸的含量,通过RT-PCR检测皮质中D1、D2两个亚型的表达。结果模型组、针刺组治疗前与空白组、假手术组比较肌张力明显增高(P <0. 01);针刺组治疗后与模型组比较,肌张力明显降低(P <0. 01);针刺组治疗前后肌张力比较差异有统计学意义(P <0. 01)。模型组与空白组、假手术组比较,大鼠大脑皮质DRD1、DRD2含量均明显降低(P <0. 01);针刺组与模型组比较,大鼠大脑皮质DRD1、DRD2含量明显升高(P <0. 05)。模型组与空白组、假手术组比较,大鼠大脑皮质DRD1mRNA、DRD2mRNA表达明显降低(P <0. 01);针刺组与模型组大鼠大脑皮质DRD1mRNA、DRD2mRNA表达比较明显增加(P <0. 05)。结论通过针刺可调节大脑皮质内DA及其受体的含量和表达,从而证明了针灸对脑卒中痉挛状态具有良好的治疗效果。
        Objective To explore the effect mechanism of acupuncture on spasticity of cerebral apoplexy(SCA through analyzing the levels of dopamine receptor and its subtypes and the expressions in the cerebral cortex in animal experiment. Methods A total of 32 SD rats were randomized into an acupuncture group(model + routine manual acupuncture at Yanglingquan GB 34,Quchi LI 11),a sham-operation group,a model group and a blank group,8 rats in each one.The cerebral middle cerebral artery suture method was used to prepare SCA. Behavioral score was adopted to determine the model success. Acupuncture started when the model was successful,lasting 3 days. ELISA was used to determine cortical dopamine(DA),DRD1 and DRD2 receptors. RT-PCR was used to determine the D1 and D2 subtype expressions. Results Before treatment,compared with the blank group and the sham-operation group,muscular tension was obviously increased in the model group and the acupuncture group(P < 0. 01). After treatment,compared with the model group,muscular tension was reduced obviously in the acupuncture group(P < 0. 01). In comparison before and after treatment,muscular tension was different significantly in the acupuncture group(P < 0. 01). Compared with the model group,the levels of cerebral cortical DRD1 and DRD2 were increased obviously in the acupuncture group(P < 0. 05). Compared with the blank group and the sham-operation group,the expressions of DRD1mRNA and DRD2mRNA in cerebral cortex were reduced obviously in the model group(P < 0. 05). Conclusion Acupuncture regulates the levels and expression of DA and its receptors in cerebral cortex. It is proved that acupuncture is effective in treatment of spasticity of cerebral apoplexy.
