摘要
目的观察肿瘤转移相关基因Mta1(metastasis associated gene 1)及其蛋白在头颈部鳞状细胞癌中的表达,并探讨其临床意义。方法对86例头颈部鳞状细胞癌组织和20例头颈部正常组织用逆转录-PCR(RT-PCR)方法检测Mta1 m RNA表达,用免疫组化检测Mta1蛋白表达。结果 Mta1 m RNA在86例头颈部鳞状细胞癌组织中平均表达水平(1.72±0.56)%与头颈部正常组织(0.93±0.68)%比较,差异具有显著性(P<0.01);Mta1蛋白在头颈部鳞状细胞癌组织阳性率(65.12%,56/86)与对照组(15%,3/20)比较,差异具有显著性(P<0.01)。Mta1 m RNA及其蛋白在下咽癌、鼻咽癌、颈部皮肤鳞癌、淋巴结癌、扁桃体癌中平均表达水平及阳性率分别与头颈部正常组织比较,差异具有显著性(P<0.01);Mta1 m RNA及蛋白在头颈部鳞状细胞癌中表达及阳性率与高分化、中分化及低分化等分化程度呈正相关(P<0.05)。结论 Mta1 m RNA及其蛋白表达与头颈部鳞状细胞癌浸润转移关系密切,提示该基因可成为预测头颈部鳞状细胞癌转移和预后有价值的指标之一。
        Objective To observe the expression of Metastasis Associated Gene(Mta1) and its protein in head and neck squamous cell carcinoma and its clinical significance. Methods Mta1 m RNA expression was detected by reverse transcription-PCR(RT-PCR) in 86 head and neck squamous cell carcinoma tissues and 20 normal head and neck tissues.The expression of Mta1 protein was detected by immunohistochemistry. Results The average expression level of Mta1 m RNA in 86 cases of head and neck squamous cell carcinoma was(1.72±0.56) compared with(0.93±0.68) in head and neck tissues. The difference was statistically significant(P<0.01). The positive rate of Mta1 protein in squamous cell carcinoma of the head and neck(65.12%, 56/86) was significantly higher than that of the control group(15%, 3/20)(P<0.01). The expression and positive rate of Mta1 m RNA and its protein in hypopharyngeal carcinoma, nasopharyngeal carcinoma, neck cutaneous squamous cell carcinoma, lymph node carcinoma, and tonsillar carcinoma were significantly different from those of normal tissues of head and neck, respectively(P<0.01). The expression and positive rate of Mta1 m RNA and protein in head and neck squamous cell carcinoma were positively correlated with the degree of differentiation of well differentiated, moderately differentiated and poorly differentiated(P<0.05). Conclusion The expression of Mta1 m RNA and its protein is closely related to the invasion and metastasis of head and neck squamous cell carcinoma, suggesting that this gene may be useful for predicting the metastasis and prognosis of head and neck squamous cell carcinoma.
