加减八珍汤对人鼻咽癌6-10B裸鼠移植瘤生长抑制作用及瘤体组织中cyclinD1表达水平影响

摘要
目的:构建人鼻咽癌6-10B裸鼠移植瘤模型,给予该模型鼠加减八珍汤处理,通过HE染色、RT-RCR及Western blot,探讨该方对鼻咽癌裸鼠移植瘤生长及增殖能力的影响。方法:将10只建模成功的鼻咽癌裸鼠随机分为实验组和对照组,分别给予加减八珍汤(浓度150 mg/mL)和0.9%生理盐水各0.2 mL灌胃,每天两次,共14 d。观察裸鼠瘤体积的变化至第21天,显微镜观察移植瘤组织病理变化特点,RT-RCR及Western blot检测移植瘤组织中cyclinD1的表达。结果:HE染色证实人鼻咽癌6-10B裸鼠移植瘤建模成功。RT-PCR和Western blot检测结果表明加减八珍汤可显著下调cyclinD1 mRNA和蛋白质的表达(P<0.05),抑制癌细胞的增殖,延缓肿瘤的生长。结论:在裸鼠体内,加减八珍汤能抑制人鼻咽癌细胞6-10B的增殖,从而达到抑制鼻咽癌组织生长的作用。
        Objective:To construct a human nasopharyngeal carcinoma 6-10 B nude mice xenograft model and to treat the model rats with addition and subtraction of Bazhen Decoction. The HE staining,RTRCR and Western blot were used to investigate the effects and mechanism on the tumor tissue in nude mice. Methods:Ten nude mice with successful modeling of nasopharyngeal carcinoma were randomly divided into experimental group and control group. They were given addition and subtraction of Bazhen Decoction(concentration 150 mg/mL)or 0.9% normal saline 0.2 mL each twice a day and 14 days in total. The tumor volumes of nude mice were observed until the 21 st day. The pathological changes of the transplanted tumor were observed under microscope. The expression of cyclinD1 was detected by RT-RCR and Western blot. Results:HE staining confirmed the successful modeling of human nasopharyngeal carcinoma 6-10 B nude mice. The results of RT-PCR and Western blotting showed that the decoction could significantly down-regulate the expression of cyclinD1(P<0.05),inhibit the proliferation of cancer cells and delay the growth of tumors. Conclusion:In nude mice,addition and subtraction of Bazhen Decoction can inhibit the proliferation of human nasopharyngeal carcinoma cell line 6-10 B,thereby inhibiting the growth of nasopharyngeal carcinoma.
