四逆酸枣仁汤合方对失眠大鼠海马GABAARα1、γ2及NKCC1的影响

摘要
目的:观察四逆酸枣仁汤合方对失眠大鼠海马GABAARα1、γ2及NKCC1的影响。方法:采用SD大鼠腹腔注射对氯苯丙氨酸(PCPA)复制失眠模型,将复制成功的模型大鼠随机分为模型组、西药组、四逆散组、酸枣仁汤组、合方组,并设置空白对照组,各组给予相应药物连续7 d,模型组和正常组生理盐水连续灌胃7 d,并观察各组大鼠的一般活动状态和体质量增量。采用实时荧光定量PCR检测海马中GABAARα1、γ2和NKCC1mRNA的表达;采用Western Blot的方法检测海马中GABAARα1、γ2和NKCC1蛋白的表达。结果:四逆酸枣仁汤合方可以提高海马中GABAARα1、γ2mRNA和蛋白的表达,降低海马中NKCC1mRNA和蛋白的表达。结论:四逆酸枣仁汤合方可以通过调节失眠大鼠海马中GABAARα1、γ2和NKCC1mRNA和蛋白的表达量来改善大鼠的睡眠节律。
        Objective: To observe the effects of Sini-Suanzaoren Formula( combined formula of Sini San and Suanzaoren Decoction) on GABAARα1,GABAARγ2 and NKCC1 of hippocampus in rats with insomnia. Methods: Insomnia model was established by PCPA injection in SD rats. Successful models were randomly divided into the model group,the western medicine group,Sini San group,Suanzaoren Decoction group,and Sini-Suanzaoren Formula group. The normal rats were taken as the blank control. After seven days of intervention,the general activity and body mass of the rats were observed,mRNA expressions of GABAARα1,GABAARγ2 and NKCC1 were detected by PCR assay. Protein expressions of GABAARα1,GABAARγ2 and NKCC1 were detected by Western Blot method. Results: Sini-Suanzaoren Formula could increase mRNA and protein expressions of GABAARα1 and GABAARγ2,reduce mRNA and protein expressions of NKCC1. Conclusion: Sini-Suanzaoren Formula can improve sleep rhythm by regulating mRNA and protein expressions of GABAARα1,GABAARγ2 and NKCC1 of hippocampus in rats with insomnia.
