二子益肾苦荞茶制备工艺及其含量测定

摘要
目的:研究二子益肾苦荞茶的制备工艺并对其进行含量测定。方法:以干膏得率与鞣花酸转移率为指标,在单因素试验基础上,采用正交试验考察加水量、提取时间、提取次数对二子益肾苦荞茶中枸杞子、覆盆子提取工艺的影响,对苦荞吸附枸杞子、覆盆子药液比例进行考察,确定二子益肾苦荞茶的制备方法 ;并采用HPLC测定二子益肾苦荞茶中芦丁、鞣花酸的含量。结果:枸杞子和覆盆子的最佳提取工艺为8倍量水,提取3次,每次2 h,出膏率为47.53%,鞣花酸提取率为97.76%;苦荞与枸杞子、覆盆子提取液的吸附比例为1∶0.5时吸附效果最佳;芦丁和鞣花酸的线性关系良好(r>0.999),芦丁和鞣花酸的加样回收率均值分别为103.02%(RSD=1.37%)和102.16%(RSD=1.09%)。结论:该试验研究的最佳提取工艺简单经济;最佳的吸附比例合理,成型工艺可行;该研究建立的含量测定方法简单、准确、灵敏、重复性好,为二子益肾苦荞茶的进一步开发、品质评价和质量控制提供了科学依据。
        Objective :To study the optimum preparation process of Erzi Yishen Buckwheat Tea and to determine its content. Methods :The optimum extraction process was determined by orthogonal experiment method with the extraction rate of extract and tannic acid as the index,the optimum molding process was determined by different adsorption ratios,and the content of tea product was determined by HPLC method. Results :The optimum extraction technology of Lycii Fructus and Rubi Fructus was 8 times water,3 times,each time for 2 hours,the extract rate was 47.53%,the extraction rate of tannic acid was 97.76%,the ratio of Tartary buckwheat to Lycii Fructus and Rubi Fructus extract was 1∶0.5,the best adsorption effect was obtainedt. The linear relationship between rutin and tannic acid is good(r>0.999). The average returning rate of rutin and tannic acid were 103.02%(RSD=1.37%)and 102.16%(RSD=1.09%). Conclusion :The optimum extraction technology is simple and economical,the best adsorption ratio is reasonable,and the molding process is feasible. The method for determination the content established in this study is simple,accurate,sensitive and reproducible,which is useful for the further development,quality evaluation and quality control of Erzi Yishen Buckwheat Tea provide scientific basis.
