摘要
目的研究白细胞介素-10(IL-10)、抗血管内皮细胞抗体(AECA)在自身免疫肺气肿大鼠中的作用。方法将人脐静脉内皮细胞向大鼠腹腔内注射建立自身免疫肺气肿模型,并设对照组,21 d建模成功,对右肺组织行HE染色病理学观察,测量肺平均内衬间隔(MLI)及平均肺泡数(MAN),对左肺灌洗并收集灌洗液(BALF),用ELISA检测BALF中的IL-10、AECA。结果与对照组比较,模型组中的MLI升高,MAN降低(P<0.05);与对照组比较,BALF中的AECA升高,IL-10降低(P<0.05)。对模型组大鼠BALF中AECA与IL-10行相关性分析,发现AECA与IL-10呈负相关(r=-0.64,P<0.05)。结论 IL-10的下调与AECA的增加共同促进了大鼠自身免疫肺气肿的形成,推测可能机制与AECA抑制IL-10的表达有关。
        Objective To study the expression of interleukin-10(IL-10) in the autoimmune emphysema in rats. Methods The model was established by human umbilical vein endothelial cells, and set up a contral groud,which was successfully modeled in 21 days.The lung tissue sections of rats were observed the morphological changes, and measured the mean linear intercept(MLI) and mean alveolar number(MAN).The left lung was lavaged to prepare BALF.The levels of IL-10 and AECA in BALF were detected by ELISA. Results Compared with the control,the MLI of the model group increased and the MAN decreased, the levels of IL-10 in BALF was lower, however, the AECA in BALF was higher. All the differences were significant(P<0.05). Correlation analysis of AECA and IL-10 in BALF of rats in the model group showed a negative correlation(r=-0.64, P<0.05). Conclusion The decrease in IL-10 and the increase of AECA concentration may have a facilitation effect in autoimmune emphysema,which may be related to the inhibition of the expression of IL-10 by AECA.
