摘要
目的通过生物信息学方法,利用GEO数据库分析鼠种植体周围袖口组织与牙齿结合上皮的差异基因表达谱。方法从GEO数据库下载芯片数据(GSE56533),运用GEO在线分析软件GEO2R对差异表达基因进行筛选,使用Cytoscape的ClueGO和CluePedia插件对差异基因进行GO及KEGG富集分析。结果筛选出722个差异表达基因(上调:455个/下调:267个)。GO富集分析结果提示差异基因在细胞间粘附、IL6的生成及白细胞趋化的生物学过程中发挥重要作用;KEGG通路结果显示差异表达基因主要在细胞因子受体相互作用、肿瘤坏死因子等信号通路上发挥作用。结论基因芯片数据分析和生物信息学的方法能从整体的角度分析种植体袖口周围软组织与正常牙龈结合上皮间的差异基因,为进一步揭示种植体周围炎机制提供新思路。
        Objective To analyze the gene expression profiles of mouse peri-implant cuff tissue and teethbinding epithelium by bioinformatics and GEO database.Methods The chip data(GSE56533) was downloaded from GEO database, the differentially expressed genes were screened by GEO online analysis software GEO2 R,and the differentially expressed genes were enriched by GO and KEGG using Cytoscape ClueGO and CluePedia plug-in.Results 722 differentially expressed genes were screened(up :455 genes/down :267 genes).The results of GO enrichment analysis indicated that the differentially expressed genes played an important role in the biological processes of intercellular adhesion, IL6 production and leukocyte chemotaxis. The results of KEGG pathway showed that differentially expressed genes mainly played a role in cytokine receptor interaction, tumor necrosis factor and other signaling pathways.Conclusion The microarray data analysis and bioinformatics method can analyze the genes between the soft tissue around the implant cuff and the normal gingiva junctional epithelium from a macroscopic and holistic perspective, providing new ideas for further revealing the mechanism of peri-implant inflammation.
