三甲基吡嗪及四甲基吡嗪对急性酒精肝损伤小鼠的保护作用

摘要
目的探讨三甲基吡嗪(Pyr)、四甲基吡嗪(TMP)及其联合作用对急性酒精肝损伤小鼠的保护作用。方法60只ICR小鼠随机分为5组,分别为对照组、模型组、Pyr组、TMP组及Pyr+TMP组。对照组自由喂养,模型组每日给予0.9%的生理盐水灌胃,其他各组分别给予100 mg/(kg·d)bw的Pyr、TMP及Pyr+TMP灌胃。30d后,造急性酒精肝损伤模型,常规方法制备小鼠血清及肝匀浆,测定血清中总胆红素(TB)、谷草转氨酶(sGOT)、谷丙转氨酶(sGPT)、碱性磷酸酶(sALP)、血糖、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)、乙醇脱氢酶(ADH)及肝匀浆中丙二醛(MDA)、谷胱甘肽(GSH)含量。结果(1)提高ADH活性:Pyr+TMP组>TMP组>Pyr组;(2)降低因急性酒精肝损伤引起的sGOT和sGPT:Pyr组>Pyr+TMP组>TMP组;(3)降低血糖和血脂,抗脂质过氧化活性:Pyr+TMP组>Pyr组>TMP组。结论 100 mg/(kg·d)bw的Pyr、TMP及两者联合使用对于急性酒精肝损伤小鼠均具有很好的保护作用,且Pyr和TMP联合使用效果最佳。
        Objective To study the protective effects of 2,3,5-trimethylpyrazine(Pyr) and 2,3,5,6-tetra methylpyrazine(TMP) on acute alcoholic liver injury in mice. Methods Sixty healthy mice rats were randomly divided into five groups:control group, model group, Pyr group, TMP group and Pyr+ TMP group. The mice were given saline or 100 mg/(kg·d) bw drug for 30 d, 12 ml/kg volume fraction of 50% ethanol were given to all groups except the control. The level of total bilirubin(TB), glutamate-oxalacetate transaminease(sGOT), glutamate pyruvate transaminase(sGPT), alkaline phosphatase(sALP), blood glucose, total cholesterol(TC), total triglycerides(TG), high-density lipoprotein(HDL-C), low-density lipoprotein(LDL-C), alcohol dehydrogenase(ADH) in serum and the content of glutathione(GSH), malondialdehyde(MDA)in liver were detected. Results(1)Improvement of the ADH activity: Pyr+TMP > TMP >Pyr;(2)Reduction of sGOT and sGPT: Pyr >Pyr+TMP > TMP;(3)Reduction of the level of glucose and serum lipid, and anti-lipid peroxidation activity:Pyr+TMP > Pyr > TMP. Conclusion Pyr and TMP(100 mg/kg bw·d) had an effective protection against acute alcoholic liver injury in mice and Pyr+TMP was the best.
