摘要
目的研究玉屏风散对人皮肤成纤维细胞光老化的保护作用及其作用机制。方法紫外线(UVA+UVB)照射人皮肤成纤维细胞,建立光老化模型,使用三种不同浓度的玉屏风散含药血清进行干预,通过β-半乳糖苷酶染色法检测细胞的衰老情况,ELISA法检测细胞DNA总体甲基化水平,透射电镜观察细胞超微结构的变化。结果与正常组相比,模型组细胞β-半乳糖苷酶染色结果阳性率明显上升,DNA总体甲基化水平降低,细胞形态发生明显改变,胞浆中可见大量空泡形成,线粒体显著肿胀变形,形态不规则。经玉屏风散干预后,人皮肤成纤维细胞的衰老程度有所减轻,β-半乳糖苷酶染色结果细胞阳性率明显下降,细胞的DNA总体甲基化水平有所升高,对成纤维细胞内细胞器尤其是线粒体的破坏明显减轻,细胞形态也有明显的改善。结论玉屏风散可能通过调控细胞DNA总体甲基化水平,保护细胞的形态,进而对于人皮肤成纤维细胞的光老化具有防护作用,其中又以大剂量玉屏风散的抗衰效果为佳。
        Objective To study the protective effect of Yupingfeng Powder on photoaging of human skin fibroblasts and its mechanism.Methods Ultraviolet(UVA+UVB)irradiation in human skin fibroblasts was used to establish a photoaging model.Three different doses of serums containing Yupingfeng Powder were used for intervention.Cell senescence was detected byβ-galactosidase staining Rit,total DNA methylation was detected by ELISA method,and cell ultrastructural changes were observed by transmission electron microscopy.Results Compared with the normal group,the positive rate of cells resulted fromβ-galactosidase staining in the model group was significantly increased,the overall DNA methylation level was decreased,the cell morphology was significantly changed,a large number of vacuoles were formed in the cytoplasm,and the mitochondria were markedly swollen and deformed with irregular morphology.After the intervention of Yupingfeng Powder,the aging degree of human skin fibroblasts was reduced,the positive rate of cells resulted fromβ-galactosidase staining was significantly decreased,the overall DNA methylation level was increased.The damage of organelles,especially mitochondria in fibroblasts was significantly reduced,and the cell morphology was also significantly improved.Conclusion Yupingfeng Powder may protect the morphology of cells by regulating the DNA methylation level of cells,and thus has a protective effect on the photoaging of human skin fibroblasts,among which the anti-aging effect of large dose of Yupingfeng Powder is the best.
