DTAF柱前荧光衍生高效液相色谱法检测食品中的克百威

摘要
本文以5-(4,6-二氯三嗪基)氨基荧光素(DTAF)~([1])为衍生试剂与克百威(CAR)发生柱前衍生反应,使其带有荧光基团并增强荧光性能,然后利用高效液相色谱法结合荧光检测器(HPLC-FLD)对其进行检测。优化了克百威的水解时间、温度等衍生反应条件和色谱分离条件。图1是(a)克百威标准溶液和(b)空白溶液分别与DTAF衍生反应之后得到的产物(CAR-DTAF)的色谱图。最终得到克百威的检出限为0.20μg/L,采用固相萃取法~([2])对四种食品样品(黄瓜、白菜、苹果、脐橙)中的克百威进行了富集提取,加标回收率范围为88.00%~105.21%。
The reaction of 5-(4,6-dichlorotriazinyl) aminofluorescein(DTAF) with carbofuran have been studied by HPLC-FLD method.In order to improve the sensitivity,the hydrolysis time and temperature of carbofuran,derivatization reaction conditions such as time,pH and chromatographic separation conditions were optimized.The detection limit of carbofuran was 0.20 μg/L under optimum conditions.A solid-phase extraction(SPE) method was used to extract four samples of vegetables and fruits before derivatization and the spiked recovery was88.00%~105.21%for carbofuran.
