GCRV免疫草鱼母本对子代早期发育中IgM、LSZ和C_3的表达影响

摘要
为探究体液免疫因子IgM、LSZ、C_3在草鱼代间的传递机制和免疫时效,本实验以草鱼出血病弱毒疫苗免疫产前40d草鱼母本,检测母本血液与子代早期发育阶段中3种免疫因子的基因表达和蛋白活性。Real-Time PCR结果表明,疫苗免疫后,母本血液中IgM、LSZ、C_3基因表达显著高于对照组(P<0.05)。实验组卵子中IgM(是对照组的2.4倍)与LSZ基因(是对照组的3.1倍)在卵子-12h(原肠胚期)表达水平急剧下降,24h(器官形成期)-3d(水花)检测不到表达,而C_3基因在24h(器官形成期)才开始出现表达。ELISA等结果表明,母本血液中3种免疫因子蛋白活性表达显著高于对照组(P<0.05),在胚胎和仔鱼期蛋白活性呈先下降后升高趋势,实验组IgM抗体水平在各发育时期显著高于对照组(P<0.05);LSZ和C_3蛋白活性在3d(水花)以后,与对照组无显著差异。2月龄鱼种实验组IgM基因在免疫组织(脾、肾、头肾)表达量均显著上调。研究显示,通过GCRV疫苗免疫能提高草鱼母源性免疫因子由亲本向子代传递能力,并对子代早期发育起免疫保护作用。
In order to explore the transfer mechanismand chronergyof humoral immune factors IgM, LSZ and C_3 between generations in Grass Carp,we vaccinated the broodstocks of the fish which was 40 d before antenatal with GCRV attenuated vaccine todetect gene expression and protein activityof parental blood and early developmental stages. The results of Real-Time PCR showed Grass Carp maternal blood IgM, LSZ and C_3 gene expression was significantly higher than the control group(P<0.05)after vaccination. The level of IgM geneand LSZ gene's expression during gastrulation was decreased rapidly,organ formation period-Fry was not detected expression,C_3 gene's expression duringorgan formation period began appearing. The ELISA indicated maternal blood 3 immune factors protein expression was significantly higher than the control group(P<0.05), The activities of protein in embryos and larvae Showed a trend from decline to rise.
