UPLC-MS/MS同时测定水产品中7种生物胺

生物胺是一类低分子量含氮有机化合物,大多由微生物来源的氨基酸脱羧酶对氨基酸脱羧后形成,常存在食品尤其是富含蛋白质的水产品中。水产品风味独特、营养丰富,备受消费者喜爱,生物胺的存在不仅会影响着水产品的品质,还会危害人类的健康,引起过敏性反应。因此,建立生物胺的检测方法对监控水产品的品质与安全至关重要。本文利用超高效液相-串联质谱(UPLC-MS/MS)建立一种无需衍生同时检测水产品中7种生物胺(色胺、2-苯乙胺、酪胺、三甲胺、组胺、尸胺、腐胺)的分析方法。通过优化确定了样品经0.2%甲酸乙腈-水(80+20,V/V)溶液提取,用Oasis Prime HLB通过流程净化;采用乙腈和10mmol/L甲酸铵(甲酸调pH至3.0)梯度洗脱,Amide色谱柱分离,电喷雾电离正离子模式(ESI+)、多反应监测(MRM)测定,外标法定量。结果显示,7种生物胺的保留时间(依次为2.41、2.43、3.19、3.40、8.22、8.24、8.42 min)均小于10 min,在各自的范围内线性关系良好,R~2>0.990;方法的检出限(LOD)为0.104~11.6μg/kg,定量限(LOQ)为0.343~38.28μg/kg。结果表明,本研究建立的UPLC-MS/MS方法简单、快速,适用于水产品中多种生物胺的同时检测。
Biogenic amines(BAs) are low molecular weight organic nitrogenous compounds bases.BAs can be found in many foods especially in seafood which are full of protein and amino acid and are mainly produced by microbial decarboxylation of amino acids.Seafood is very popular among people due to its unique flavor and rich nutrition,however,the existence of BAs would not only decrease their quality value,but also have a bad effect on human health such as allergic reactions.Thus,it is essential to establish analysis method of BAs in aquatic product for monitor their quality and safety.This dissertation has been developed a no derivative method for simultaneous analysis of 7 kinds of BAs(Tryptamine,2-Phenylethylamine,Tyramine,Trimetilamine,Histamme,Cadaverine,and Putrescine) in aquatic product by HPLC-MS/MS(ultrahigh-performance liquid chromatography tandem mass spectrometry).Sample pretreatment conditions and HPLC-MS/MS conditions were optimized.Sample was extracted with 0.2%.HAc Acetonitrile-water(80 +20,V/V) directly,and purified by Oasis Prime HLB with the pass method,Acetonitrile and 10mmol/L ammonium formate(pH =3.0 adjusted with formic acid) was chose to be mobile phase with gradient elution,and then analyzed by using HPLC-MS/MS in multiple reaction monitoring(MRM) mode via positive electrospray ionization(ESI +) with an Amide column(1.7μm,2.1mm x 100 mm) as the analytical column,and the quantization was carried out by the external standard method.The chromatographic peaks of 7 kinds of BAs were flowed out within 10 minutes(2.41,2.43,3.19,3.40,8.22,8.24,8.42 min).For the above method,the linear correlation coefficients for 7 kinds of BAs were not less than 0.990 in their linear scope with good linearity.Respectively,the limit of detection(LOD) ranged from 0.343μg/kg to 11.600μg/kg.The limit of quantitation(LOQ) was in the range of 0.343 ~38.380μg/kg.The results showed that the method is simple,rapid to meet the requirements of BAs analysis.This method was applicable to simultaneous confirm the BAs in aquatic product.
