纳米水凝胶负载基于BODIPY的荧光探针用于钙离子检测

摘要
钙离子在生命体系中起着重要的作用。生物体内钙离子含量与多种生物过程密切相关,如神经兴奋、神经递质的释放、基因转录、细胞增殖和激素分泌等。使用荧光法检测钙离子,具有灵敏度高、响应迅速、易于可视化检测等优点,且可用于生物体成像,因此受到广泛关注。本工作设计了一种基于BODIPY衍生物荧光团和钙离子特异基团1,2-双(2-氨基苯氧基)乙烷-N,N,N',N'-四乙酸(BAPTA)的油溶性分子,该分子与钙离子反应之后,光致电子转移效应消失,原本被猝灭的发光重新显现,故可作为荧光发光型钙离子探针。使用纳米水凝胶负载该分子制备荧光纳米器件,可灵敏、高效、准确地检测水体系中的钙离子含量。
Calcium ions play vital role in organism. The level of calcium ions is sensitive to many biological processes, such as neural excitation, release of neurotransmitter and cell proliferation. Herein, a fluorescent probe for calcium ion detection was reported. The probe, which consisted of a BODIPY-derived fluorophore and a calcium-specific 1,2-bis(O-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid(BAPTA) moiety, could recuperate its fluorescence upon the calcium ions addition because the photon-induced electron transfer process was exhibited. After incorporated with nanogel, the water-insoluble probe could be applied in aqueous system with fast response, high sensitivity and selectivity to calcium ions.
