光谱法结合化学计量学研究孔雀石绿及其代谢产物与BSA的作用

摘要
孔雀石绿及其代谢产物(无色孔雀石绿)能迅速在组织中蓄积~([1]),无色孔雀石绿具有高残留和致癌、致畸、致突变等副作用,很多国家已经禁用,但仍有不法分子使用无色孔雀石绿延长鱼类的存活时间~([2])。本文采用经典方程Stern-volmer,双对数,Van't Hoff等方程计算作用体系的常数和参数;用红外和圆二色谱证实BSA构象的改变;并用MCR-ALS对生命体系测量的多维波谱数据进行解析和描述(Fig.1)。从结果可知:孔雀石绿及无色孔雀石绿与BSA结合常数达到10~6 L mol~(-1),血清结合率高,在体内消除慢,作用维持时间长,不易代谢;通过对荧光和圆二色谱两种光谱方法的数据矩阵进行扩展并用MCR-ALS处理数据,较全面地分析了无色孔雀石绿与BSA相互作用过程,从而得知无色孔雀石绿在内脏和组织中形成严重的蓄积性残留,危害食品安全和人类健康。
The effect of malachite green and leucomalachite green(leuco),with bovine serum albumin(BSA) has been studied by intrinsic fluorescence spectroscopy(FLS),circular dichroism(CD) and chemometric methods- multivariate curve resolution-alternating least squares(MCR-ALS).It was found that the binding constant of malachite green and leucomalachite green to BSA was 10~6 L mol~(-1).Quantitative and quanlitative information about the complex leucoprotein species formed was obtained with the resolution of the FLS and CD data matrices by MCR-ALS.This research has implications for the analysis of leuco in food as conducted e.g.by the food and drug administration(FDA) and for the preventing of risks to human health and food safety.
