龙须菜硫酸多糖的抗过敏作用研究

摘要
目的:探讨龙须菜硫酸多糖(sulfated polysaccharide from Gracilaria lemaneiformis,GLSP)在抗原刺激的小鼠模型和细胞模型中的抗过敏作用。方法:以龙须菜为原料,通过热水浸提、旋转蒸发、乙醇沉淀得到粗多糖,进一步采用DEAE+cellulose 52层析柱纯化得到GLSP。利用甲壳类主要过敏原原肌球蛋白(TM)对Balb/c小鼠进行腹腔注射和灌胃,建立过敏反应小鼠模型,以及通过RBL-2H3和KU812细胞模型,研究GLSP的抗过敏作用;通过蛋白质组学方法研究GLSP的抗过敏作用机理。结果:经DEAE+cellulose 52层析柱纯化得到152 kDa含11.26%硫酸根的GLSP。GLSP能够降低小鼠血清中TM特异性的IgE和IgG1,抑制小鼠肥大细胞释放肥大细胞蛋白酶和组胺,从而显著缓解TM致敏的小鼠过敏反应症状,并能够抑制Th2细胞极化,促进调节性T细胞功能。通过RBL-2H3细胞实验得知,GLSP能够显著抑制过敏标记物,包括β-氨基己糖苷酶、组胺、活性氧和过敏反应相关的细胞因子(IL-4和TNF+α);同时GLSP能够降低KU812细胞表面CD63和CD203c的表达;最后发现GLSP主要通过抑制Syk和MEK3/6+p38MAPK信号通路起到影响KU812细胞激活的作用。讨论:GLSP通过免疫抑制作用缓解小鼠过敏反应症状,通过抑制Syk和p38 MAPK信号通路抑制过敏反应效应细胞的激活,从而可知GLSP可以用于预防或治疗过敏反应疾病。
The present study investigated the anti-allergic potential of the sulfated polysaccharide from Gracilaria lemaneiformis(GLSP) in antigen-sensitized mice and cells.The polysaccharides which were 152,481 Da composed of 11.3%sulfate were isolated from Gracilaria lemaneiformis with hot-water extraction,evaporated under reduced pressure,precipitated by ethanol,and purified by DEAE-cellulose 52 chromatographic column orderly.The anti-allergic activity was investigated by TM-sensitized Balb/c mice model,RBL-2H3 cells and KU812 cells.The results demonstrated that GLSP could alleviate allergy symptoms,effectively reduce TM-specific IgE and IgGl,decrease the levels of histamine and mMCP-1,suppress Th2 cell polarization,and promote the function of regulatory T cells.In addition,GLSP had the ability to inhibit the production of(3-hexosaminidase,histamine,ROS,IL-4 and TNF-α in IgE-mediated RBL-2H3 cells.Meanwhile,the expression of CD63 and CD203 c were reduced by GLSP in activated KU812 cells.GLSP inhibited the activation of KU812 via suppressing Syk and p38mitogen-activated protein kinases(MAPK).Taken together,immunosuppression as well as the reduction of Syk and MAPK p38 may contribute to GLSP persistent protection against food allergy.The results suggest that the sulfated polysaccharide from Gracilaria lemaneiformis may provide insight into the prevention and treatment of allergic diseases.
