银杏内生菌Endo Gin Ya1胞外多糖发酵条件的优化

摘要
银杏多糖具有较强的免疫调节抗肿瘤、抗氧化、抑菌等生物学功能,但银杏资源的有限性限制了对银杏多糖的开发和利用。植物内生菌能够产生与宿主相同或相似的生理活性成分,本实验利用银杏内生Endo Gin Ya1生产胞外多糖,并利用单因素正交试验,从培养基和培养条件两方面对其发酵条件进行优化,从而确定最佳发酵条件:培养基组成为蔗糖16%,酵母浸粉1.0%,氯化钠0.5%,氯化锰0.1%,氯化镁0.1%,氯化钙0.1%,培养基初始pH6.5;发酵温度26℃,装液量70mL,接菌量1.0%,摇床转速160 rpm,培养时间48h,在此条件下菌株胞外多糖产量最高,可达8.72异/L。通过对发酵条件的优化,大大提高了银杏内生菌发酵生产胞外多糖的产量,为以后的深入研究和其在食品和制药行业的应用奠定了基础。
Ginkgo polysaccharide has strong biological function,such as immune regulation,anti-tumor,anti oxidation,anti bacteria and so on.However,the limitation of the ginkgo resources restricts the development and utilization of Ginkgo polysaccharide.Endophytic bacteria can produce the same or similar physiological active components as the host.Ginkgo biloba endophyte Endo Gin Yal was used to product extracellular polysaccharide depending on single factor and orthogonal experiments.Culture conditions were determinedas followed:fermentation medium was composed of 16%sucrose,1.0%yeast extract,0.5%sodium chloride,manganese chloride 0.1%,0.1%magnesium chloride and calcium chloride 0.1%at initial pH6.5,26℃ with 70 mL in 250 mL shaking flasks,and with inoculation amount 1.0%at 160 rpm for 48 h.Under the optimal conditions,the strain of extracellular polysaccharide yield reached 8.72g/L.Through the optimization of fermentation conditions,the production of extracellular polysaccharide from fermentation of Ginkgo biloba from Endophytic bacteria has been improved greatly,which lays a foundation for further research and its application in food and pharmaceutical industry.
