摘要
牛初乳中含有多种具有重要生理活性的寡糖,但是由于对牛初乳中寡糖的研究较少且缺乏有效分离提取方法,导致牛初乳资源极大浪费。本研究是以光明牧场牛初乳为原料,采用膜技术对牛初乳中的功能性寡糖进行初步分离,构建出一种新型树脂介质进一步分离纯化,并利用液质联用方法对纯化得到的寡糖进行结构鉴定。结果表明:10kDa超滤膜可以去除98.9%的蛋白质且总糖的回收率为96.7%;利用500Da纳滤膜去除了部分乳糖及单糖,最后得到的寡糖混合物中功能性寡糖占总合量的44%;通过液质联用鉴定出10种寡糖,其中唾液酸乳糖含量最高为1.7g/l,是牛初乳中除了乳糖外含量最高的一种功能性寡糖;构建的新型树脂对牛初乳中的唾液酸化寡糖具有较好的分离效果,同时由于其制备过程简单、成本低廉,具有大规模化分离纯化牛初乳中唾液酸化寡糖的潜力。
Bovine colostrum contains many functional oligosaccharides,but the utilization of them was low because of less research and no effective method to separation and purification.This study using membrane technology in preliminary separation,then developed a new Chromatographic packing for its further purification,use Liquid Chromatography-Mass Spectrometry to qualitative analysis of the oligosaccharides.The results show that thelOkDa ultrafiltration membrane were remove 98.9%of protein and obtained 96.7%of total sugar;500Da nanofiltration membrane to separate lactose and monosaccharide,finally obtained 44%of functional Oligosaccharides in the mixed oligosaccharide,10 oligosaccharides in the isolate was identified by LCMS and in it sialyllactose was about 1.7g/l,which was the most abundant oligosaccharides in bovine colostrum except lactose.The new material 1 have high separation efficiency for sialy-oligosacharides,due to its low cost,easer to produce it has potential to separation and purification sialy-oligosacharides in large-scale.
