自噬对小鼠胚胎成纤维细胞饥饿条件下的脂类代谢影响研究

摘要
自噬是真核细胞内普遍存在的一种生理现象,越来越多的研究表明自噬与人类疾病有着密切的联系。Atg7蛋白在自噬小体的形成中起着重要的催化作用,对于自噬过程不可或缺。本研究利用正相/反相二维高效液相色谱-质谱联用(NP/RP2DHPLC-MS)技术,分析野生型(wild-type)及Atg7基因敲除型(Atg7~(-/-))小鼠胚胎成纤维细胞(MEFs)饥饿条件下,其胞内脂类的变化情况。饥饿时间设为0、2、4小时,并将饥饿后细胞换回正常培养基4小时作为恢复组。共在wild-type MEFs和Atg7~(-/-)MEFs中分别检测到492和486种脂质分子,其中212和231种在饥饿过程中含量发生了1.5倍以上变化,包括鞘脂类,甘油脂类和脂肪酸类。分析两种细胞中脂类分子的变化趋势,在wild-type MEFs中,脂类分子大多呈现先上升后逐渐降低的趋势,且恢复组含量能回到未饥饿时水平;而在Atg7~(-/-)MEFs中,绝大多数脂类分子在饥饿发生后呈现一直下降的趋势,且多数未能恢复正常水平。表明自噬过程的发生影响了MEF的脂类代谢,进一步的研究将会从脂类转化通路方面,给出更具体的生物学解释和验证。
Autophagy is a ubiquitous physiological phenomenon in eukaryotic cells,and more and more researches revealed that it was closely connected with various human diseases.Atg7 plays an important role in the formation of autophagosome,and is indispensable in the induction of autophagy.In this study,a NP/RP 2D HPLC-MS method was employed to analyze the lipid metabolism of wild-type and Atg7~(-/-) MEFs in response to nutrient stress.The starvation time was set as 0 hour,2 hours and 4 hours as well as the recovery group(cultured back in full medium).492 and 486 lipid species were successfully detected in wild-type and Atg7~(-/-) MEFs respectively,among which 212 and 231 were found to be altered with fold change more than 1.5 during the starvation time.The trend of these lipids were quite different in the two cell lines indicating that autophagy regulates lipid metabolism of MEFs in response to starvation.
