基于亚细胞水平的拉曼光谱及成像技术的结肠癌诊断研究

摘要
组织细胞发生癌变过程中,细胞内的生物分子构型、构象及各成分的构成比例会发生明显变化。共聚焦显微拉曼光谱技术能够捕捉分子水平的变化差异,并将此差异反映在拉曼光谱上。本研究采集正常细胞和癌细胞的拉曼图像,结合经典最小二乘法(CLS)模型对图像信息进行分析,证明正常细胞与癌细胞中细胞核与细胞质存在分布及光谱信息的差异。因此分别采集人体实际组织正常细胞与癌细胞的细胞核及细胞质的拉曼光谱,结合偏最小二乘判别分析(PLS-DA)模型对光谱信息进行分析,以细胞核与细胞质光谱的差异作为标记,对结肠癌判别的准确性可达95%以上。本研究说明,亚细胞结构的拉曼光谱分析能够显著提高对结肠癌的诊断,同时,进一步证明共聚焦显微拉曼光谱仪在癌症诊断分析领域具有巨大的应用潜力。
Cancer cells are continually undergoing transformation to enable survival,replication and metastasis.Confocal Raman microspectroscopy studies can provide a view of the biochemical changes accompanying cellular transformations.We obtained Raman spectra and image of nucleus and cytoplasm corresponding to the pathological types of colorectal tissues.Partial least-squares-discriminant analysis(PLS-DA) and classical least-squares(CLS) model,were employed to analysis the Raman spectra and image data.This work demonstrates that the confocal Raman spectroscopy has the potential to be a powerful tool for diagnosis and detection of colorectal cancer during clinical examination.
