致病性与非致病性副溶血弧菌在不同温度条件下生物被膜形成过程研究

副溶血性弧菌是水产品中重要的食源性致病菌,易形成生物被膜而使其对外界环境的抵抗力显著增强。本实验采用结晶紫染色法和激光共聚焦扫描显微镜探究了致病性(22株)与非致病性(17株)副溶血性弧菌生物被膜在不同温度(15℃,25℃和37℃)下的动态形成过程。结果表明,25℃是生物被膜形成的最适温度,主要是因为25℃时细菌鞭毛运动性强,促进了胞外多糖等物质的分泌而形成大量的生物被膜。随着培养时间的延长,15℃时生物被膜一直增加,25℃时先增加后降低且在12 h达到最大,37℃时无明显变化。此外,致病性菌株生物被膜形成能力显著强于非致病性菌株(p<0.05)。通过变异系数(CV=标准偏差/平均值×100%)分析发现,菌株的变异性随着温度的增加而增加,且均在12 h时达到最大值。本研究表明副溶血性弧菌生物被膜的形成过程在不同温度下存在差异,为控制生物被膜的危害提供了理论依据。
Vibrio parahaemolyticus is an important food-borne pathogen in seafoods,it can form biofilm easily which is more resistant to external environments.The present study investigated the dynamic process of biofilm formation of pathogenic(22strains) and non-pathogenic(17 strains) V.parahaemolyticus on polystyrene microtiter plates at different temperatures(15℃,25℃ and 37℃),using the crystal violet staining and eonfocal laser scanning microscopy(CLSM).The results indicated that 25℃ is the optimal temperature for biofilm formation,mainly due to the strong motility of bacteria flagella promoted the secretion of exopolysaccharides and extracelluar protein,and then form more biofilm at 25℃ Averaged over all strains,biofilm developmental process at 15℃ and 25℃ were divergent,biofilm formation increased continuously at 15℃,while at25℃ biofilm formation increased gradually and peaked at 12 h.However,biofilm increment was not significant at 37℃.Additionally,the biofilm formation ability of pathogenic strains,on average,was significandy stronger(p < 0.05) than nonpathogenic strains at all temperatures measured.Moreover,extensive strain variability was observed during biofilm formation and indexed using the coefficient of variation(CV = standard deviation / mean * 100%).The strain variability increased with increasing temperature and this index,at all temperatures,peaked after 12 h.The results of this study demonstrated that the developmental process of biofilm of V.parahaemolyticus was obvious difference at three temperatures,which provided a basis to control the hazards of V.parahaemolyticus biofilm.
